人肠生长激素工程菌高效表达培养工艺的研究
Studies on Cultural Technology of High Expression of the Recombinant Human Gut Growth Hormone in E.coli BL21
【摘要】 为了优化含有人肠生长激素 (rh[Gly2 ]GLP 2 )基因的融合表达工程菌 pED h[Gly2 ]GLP 2高效表达的最适培养工艺条件 ,文章通过摇瓶培养研究了包括培养基组成、初始 pH值、接种量、诱导剂 ,诱导剂浓度、诱导时机及诱导后培养时间等因素对蛋白表达量的影响 ,确定了该工程菌表达目的蛋白的最佳条件 ,目的蛋白表达量可达 4 0 %以上 ,为基因工程产品的大规模工业化生产提供了一些有价值的数据
【Abstract】 To optimize the cultural technology of the Engineering Bacteria pED-PP-h[Gly 2]GLP-2 containing Recombinant Human Gut Growth Hormone rh[Gly 2]GLP-2, Fermentation conditions, which influence the final yield of protein expression, including growth medium, original pH of culture, inoculation ratio, inducer,concentration of inducer lactose and induced time, culture time after induced and so on, were studied in shaking flasks. The results indicated that, with YM-1 medium(10 g/L monosodium glutamate, 15 g/L beef extract, 25 g/L corn extract), original pH 6.6 of culture, 2% inoculation ratio, cultured 4 hours at 37℃, and then induced 8 hours using 5 mmol/L lactose,examined by SDS-PAGE and gel scanning, the expression level of protein was over 40%.
