端粒酶hTERT蛋白与基因在人肿瘤细胞中表达的相关分析

【摘要】 目的分析端粒酶hTERT蛋白在人肿瘤细胞中的表达意义,探讨hTERT蛋白与基因在人肿瘤细胞中表达的相关性,预测其成为肿瘤诊断标志物的趋势。方法采用免疫组织化学及原位杂交方法分别对171例恶性肿瘤,61例癌前病变及良性病变进行hTERT蛋白和基因检测,同时与p53蛋白进行比较。结果hTERT蛋白及基因在恶性肿瘤中平均检出率分别为84.8%(145/171),83.4%(143/171);淋巴结转移性癌中100%(5/5),80%(45);癌前与良性病变分别为37.5%、25%的阳性,高出p53的表达(恶性肿瘤45.6%,淋巴结转移性癌53.3%,癌前病变与良性病变均为0)。结论hTERT蛋白与基因表达比较一致,均与肿瘤的恶性行为有密切关系,明显高于p53且免疫组织化学操作简单易行,提示端粒酶hTERT蛋白有可能是一个比p53更好的肿瘤诊断的新标记物.

【Abstract】 Objective To get insight into the expression of telomerase hTERT protein associated with hTERT gene in human tumors,and to compare with accumulation of p53to determine its value in cancer clinical diag-nosis.Methods One hundred and seventy-one cases of cancers,sixty one cases of precarcinomatous lesions or benign ones were analyzed for hTERT,p53protein expression,and hTERT gene alteration by immumohistochemistry.and in situ hybridization Results hTERT protein and gene positive rates in cancers were averagely84.8%,83.4%;in metasti-atc carcinomas reached100%,80%respectively.The precancerous lesions and benign lesions were positive in37.5%,25%.The positive rate of p53in cancers was about45.6%;in metastatic carcinomas reached60%(3/5).p53expresson of precancerous lesions and benign lesions was all negative.Conclusion The expression of telomerase hTERT protein and gene correlated closely with malignant phenotype of tumors,and was more frequently positive than p53,suggesting that hTERT might be a new indicator to carcinoma’s diagnosis.
