高效液相色谱手性流动相法拆分帕珠沙星对映体
Study on Enantiomeric Separation of Pazufloxacin by HPLC
【摘要】 目的:对帕珠沙星对映体进行手性分离。方法:采用高效液相色谱手性流动相法,用ODS柱(4.6 mm×150 mm,10 μm),检测波长320 nm,流动相为甲醇-手性溶液(25:75,pH 3.0),流速1 mL·min-1。结果:右旋体最低检测限1.5 ng,重现性好。结论:该法可用于帕珠沙星光学纯度检查。
【Abstract】 Objective: Study for the chiral separation of the enantiomeric in pazufloxacin. Methods: Enantiomer of pazufloxacin was determined by HPLC with a chiral mobile phase additive ( CMPA). C18 column (4. 6 mm × 150 mm, 10 μm) was used. The mobile phase consisted of methanol - chiral liquid(25:75 ,pH 3. 0). Flow rate was 1 mL· min-1. UV wavelength was 320 nm. Results:The detection limit was 1. 5 ng for enantiomer of pazufloxacin. The reproducibility has been found to be RSD 0. 41% . Conclusion: The method can be used for determination of enantiomeric purity for pazufloxacin.
