1株嗜碱克氏菌属菌株的分离及系统发育分析
Isolation and phylogenetic analysis of an alkaliphilic strain of the genus Kocuria
【摘要】 从埃及盐碱土样分离得到1株革兰氏阳性的兼性嗜碱菌株YIM70003,不产荚膜和芽孢,不具游动性.生长温度范围为室温(约20℃)到37℃,最适生长温度为28℃,生长的pH值范围为pH5~12,对NaCl的耐受范围为1%~5%,DNA的G+Cmol%为69.44%.16SrRNA全序列分析的结果表明,该菌株与Kocuriapo larisCMS76T亲缘关系最近,但两者在形态、生理生化特征上有差异,有可能是克氏菌属的一个潜在新种.
【Abstract】 An Gram-positive facultative alkaliphilic strains were isolated from alkaline saline soils collected from Egypt,which has the following characteristics:non-endospore-forming,non-encapsulated,non-motile.Grows between room tempreture(about 20℃)and 37℃,with optimum growth at 28℃.Grows in Horikoshi medium adjusted to pH5—12 and tolerates up to 1%—5% NaCl.The G+C content of DNA was 69.44%.Analysis of l6S rRNA gene sequences clearly indicated that the isolate was a member of the genus Kocuria.Strain YIM70003 exhibited maximum similarity with the 16S rDNA sequence of Kocuria polaris CMS 76T,while the two strains had great difference with their phenotypic and physiological characteristics.Thus strain YIM70003 maybe represent one potential new species of the genus.
