细胞间黏附分子-1在糖尿病大鼠视网膜中的表达及意义
Expression and significance of intercellular adhesion molecule-1 in retin a of diabetic rats
【摘要】 目的探讨细胞间黏附分子-1(ICAM-1)在糖尿病大鼠视网膜中的表达及ICAM-1多克隆抗体(ICAM-1Pab)的治疗作用。方法Wistar大鼠40只随机分成4组正常对照组(CON)、糖尿病组(DM)、糖尿病(ICAM-1PAb)组(DM(ICAM-1PAb))和糖尿病(生理盐水)组(DM(saline)),每组10只。大鼠腹腔注射链脲佐菌素(STZ)诱发糖尿病模型。DM(ICAM-1PAb)组右眼结膜下注射ICAM-1PAb,连续1周。制作大鼠视网膜冰冻及石蜡切片分别行免疫组化SP染色和常规HE染色,对SP染色结果作计算机图像分析。结果ICAM-1在大鼠视网膜血管内皮细胞胞膜有表达。SP染色结果采用计算机图像分析系统处理。其中DM组分别与CON组、DM(ICAM-1PAb)组比较有显著性差异(P<0.01)。DM组视网膜血管内有较多附壁的白细胞。结论早期糖尿病大鼠视网膜ICAM-1表达上调,ICAM-1PAb通过阻断ICAM-1,抑制白细胞黏附,改善糖尿病大鼠视网膜微循环。
【Abstract】 Objective To investigate the expression of intercellula r adhesion molecule-1(ICAM-1)in retina of diabetic rats and the therapeutic effect of ICAM-1polyclonal antibody(ICAM-1Pab).Methods Fourty healthy ad ult Wistar rats were chosen and randomly divided into normal control(CON),dia betes mellitus(DM),diabetes mellitus (ICAM-1PAb) (DM ICAM-1Pab ),and diabetes mellitus (saline) (DM saline )groups.Diabetes mellitus of rats was induced by injection of streptozotoc in(STZ)intraperitondally.ICAM-1PAb was subconjunctival injected for one wee k in the right eye of rats in DM ICAM-1Pab group.Retinal frozen and paraffin sections were stained by immunhistochemic al SP and HE method respectively and analyzed by computer-picture analysis syst em.Results Only retinal vascular endotheliocyte showed ICAM-1positive reactio n.The results of computer-picture analysis in CON,DM,DM ICAM-1PAb and DM saline groups were13.75±7.86,45.10±10.28,18.23±5.8 9and42.31±8.97,respectively,indicating a significant difference in DM group compared with CON or DM ICAM-1PAb groups(P<0.01).Many adhesive neutrophils were observed in retinal va sculature of rats in DM group.Conclusion The expression of ICAM-1is upregulat ed in retina of early diabetics rats.ICAM-1PAb can improve retinal microcircu lation in diabetic rats by inhibiting neutrophil adhesion.
