HSV-TK/GCV自杀基因系统对人胰腺癌细胞的作用

【摘要】 目的 运用自杀基因系统HSV TK/GCV治疗人胰腺癌细胞SW1990的实验研究 ,并证实旁观者效应。方法 通过脂质体介导的基因转移方法将含有单纯疱疹病毒胸苷激酶基因 (her pessimplexvirusthymidinekinasegene ,HSV TK)的复制缺陷型逆转录病毒载体GINaTK导入包装细胞PA317,然后利用细胞产生的假病毒颗粒感染人胰腺癌细胞系SW 1990细胞。在SW 1990 /TK细胞培养物中加入抗病毒前药GCV ,观察GCV对该细胞的杀伤作用及旁观者效应。结果 实验表明GCV对SW1990 /TK有显著的抑制及杀灭作用 ,同时表现出强烈的旁观者效应。结论 HSV TK/GCV自杀基因系统对胰腺癌细胞有生长抑制及杀伤作用 ,为临床应用提供了实验依据

【Abstract】 Objective We used HSV-TK suicide gene in combination with prodrug GCV,to study the in vitro therapeutic effect on human pancreatic cancer,as well as the bystander effect. Methods We used Lipofectin to transfer the replication defective recombinant retrovirus vector GINaTK(bearing HSV-TK gene)to package cell PA317,and made the cell produce progeny virion,which can transfer the HSV-TK gene to target cells SW1990.We then added GCV to these gene-modified cells and studied the sensitivity of the cells to GCV as well as the bystander effect.Results The gene-modified pancreatic cancer cells SW1990/TK were sensitive to the treatment of GCV compared with unmodified tumor cells,and also we observed a remarkable bystander effect.Conclusion Our data demonstrate the efficiency of HSV-TK/GCV system and the potential feasibility of suicide gene strategy for clinical therapy of human pancreatic cancer.
