青藤碱对核转录因子κB及其抑制因子IκB的影响

【摘要】 目的 探讨青藤碱对BALB/c近交纯系小鼠腹腔巨噬细胞核转录因子κB及其抑制因子IκB表达的影响 ,提供其抗炎治疗类风湿性关节炎作用机制的实验依据。方法 应用荧光标记和激光共聚焦扫描显微镜技术检测药物对LPS(Lipopolysaccharides)刺激状态下腹腔巨噬细胞NF κBp6 5 (nucleartranscriptionfactors)核移位的影响 ;采用Westernblot方法检测药物对LPS刺激作用下的腹腔巨噬细胞IκB α降解的影响。结果 高、中剂量给药组与模型对照 (control)组间NF κBp6 5核移位和IκB α降解的差异有显著性 (P <0 0 1)。结论 青藤碱的抗炎抗风湿的作用可能与其抑制NFκBp6 5核移位和IκB α降解有关。

【Abstract】 Aim To observe the effect of Sinomenine on the activation of NF-κB and the degradation of its inhibitor I-κB in peritoneal macrophages of BALB/c mice in vitro and provide experimental evidence for further evaluation of the antiinflammatory and antirheumatic effects of Sinomenine. Methods Effect of Sinomenine on the activation of NF-κB p65 in the cells was investigated by using fluorescence-labelling and laser confocal scanning microscopy; Effect of Sinomenine on the degradation of IκB-α was investigated by Western blot. Results SIN (0.25,1.25 mmol·L -1) attenuated the activation of NF-κB p65 in peritoneal macrophages of BALB/c mice induced by LPS in vitro. SIN(0.25,1.25 mmol·L -1) inhibited the degradation of I-κB-α in peritoneal macrophages of BALB/c mice induced by LPS in vitro. Conclusion SIN can partially inhibit the activation of NF-κB p65 and the degradation of IκB-α in peritoneal macrophages of BALB/c mice induced by LPS in vitro.
