罗格列酮对果糖饲养大鼠血糖血脂及血管重构的影响
Effects of rosiglitazone on blood glucose, blood lipid and vascular remodeling in rats fed with high-fructose diet
【摘要】 目的 研究罗格列酮 (Rosiglitazone)对高果糖饲料饲养SD大鼠血管重构的作用和对血糖血脂的影响。方法 高果糖饲料饲养SD大鼠 1mon ,随机分为两组 :高果糖组 ,继续喂养高果糖饲料 1mon ;高果糖罗格列酮处理组 ,喂高果糖饲料和罗格列酮 (5mg·kg-1·d-1,溶于饮水中 ) 1mon。最后 ,每组一半大鼠麻醉从心脏取空腹血测血糖、血脂、胰岛素。每组另一半取主动脉和肠系膜小动脉 ,固定 ,切片 ,HE染色 ,用病理分析系统测量主动脉和肠系膜小动脉重构指标 :内径、中膜、中膜 /内径、中膜横切面积。结果 高果糖罗格列酮处理组与高果糖组对比 ,罗格列酮降低了血压 (16 0kPavs 18 0kPa ,P <0 0 1)、胰岛素 (34 89mIU·L-1vs5 3 5 6mIU·L-1,P <0 0 1)、甘油三酯 (0 5 6mmol·L-1vs1 16mmol·L-1,P <0 0 5 ) ,升高了胰岛素敏感指数 :血糖 /胰岛素的值 (0 2 6vs 0 14 ,P <0 0 5 ) ;增加了肠系膜小动脉内径 (173 0 μmvs 10 0 0 μm ,P <0 0 1) ,减小了中膜 (18 8μmvs 2 4 5 μm ,P <0 0 5 ) ,减小了中膜 /内径(10 90 %vs 2 5 5 5 % ,P <0 0 1) ;减小了主动脉中膜 (89 0μmvs 10 9 0 μm ,P <0 0 1) ,减小了中膜 /内径 (5 74 %vs6 80 % ,P <0 0 1)。结论 罗格列酮能降低果糖饲养SD大鼠的高血压 ,?
【Abstract】 AIM To investigate the action of rosiglitazone on blood glucose,blood lipid,and the vascular remodeling in rats fed with high-fructose diet. METHODS After fed with high-fructose diet for one month,rats were randomly divided into two groups:High-fructose diet group that continued feeding with high-fructose diet for another month; High-fructose diet+rosiglitazone group fed with high-fructose diet and rosiglitazone (5 mg·kg -1·d -1, dissolved in water) for one month. At last,one half of each group were anesthetized,and the fasting blood, obtained from heart,was used to detect blood glucose,blood lipid,blood insulin. The aorta and mesenteric artery were gotten from the other half of each group, fixed in formalin,sliced and HE dyed. Pathology analysis system was used to measure the remodeling indexes of aorta and mesenteric artery:Lumen ,media,media/lumen,media cross-section area. RESULTS Comparing high-fructose diet+rosiglitazone group with high-fructose diet group, rosiglitazone decreased blood pressure(16.0 kPa vs 18.0 kPa, P<0.01), insulin(34.89 mIU·L -1 vs 53.56 mIU·L -1, P<0.01) and triglyceride(0.56 mmol·L -1 vs 1.16 mmol·L -1, P<0.05),raised insulin sensitivity index(Fasting blood glucose/Fasting serum insulin) (0.26 vs 0.14, P<0.05),increased the lumen diameter of mesenteric artery(173.0 μm vs 100.0 μm, P<0.01), decreased media(18.8 μm vs 24.5 μm, P<0.05), decreased media/lumen(10.90% vs 25.55%, P<0.01),decreased the media of aorta(89.0 μm vs 109.0 μm, P<0.01), decreased media/lumen (5.74% vs 6.80%, P<0.01). CONCLUSION Rosiglitazone can decrease blood pressure,correct glucose and lipid metabolic abnormalities, inhibit the form of aorta and mesenteric artery remodeling in SD rats fed with high fructose diet.
