乳铁蛋白在大鼠坐骨神经慢性束缚损伤模型产生镇痛作用
Analgesia effect of lactoferrin in rat chronic constriction injury (CCI) model
【摘要】 目的 研究乳铁蛋白对大鼠坐骨神经慢性束缚损伤模型的镇痛作用。方法30只成年SD大鼠,制作坐骨神经慢性束缚损伤模型,实验动物分为5组,分别腹腔注射生理盐水及乳铁蛋白30、100、300、1 000 mg/kg。用辐射热刺激法诱发鼠后腿回缩试验测定痛阈,药物的效应以最大可能效应百分数(MPE%)表示。以t检验法分析乳铁蛋白各剂量组与对照组之间的差异。结果腹腔注射乳铁蛋白剂量依赖性地延长大鼠后腿回缩潜伏期;乳铁蛋白的峰值作用时间在用药后60 min;乳铁蛋白各剂量组的MPE%与生理盐水组相比有显著性差异(P<0.01)。结论腹腔注射乳铁蛋白对大鼠坐骨神经慢性束缚损伤模型产生剂量依赖性镇痛作用。
【Abstract】 Objective To investigate the analgesic effect of lactoferrin (LF) on neuropathic pain. Methods 30 adult male Sprague - Dawley rats were operated on to form CCI models and divided into 4 test groups (30 mg/kg, 100 mg/kg, 300 mg/kg, 1 000 mg/kg of LF) and 1 saline control group) . The neuropathy was produced in the right hind paw of the rat, where the thermal hyperalgesia induced was assessed by using rat plantar heat test. The percentage of maximum possible effect (%MPE) was calculated for each rat before and after LF administration. Student’ s t - test was employed to analysis the difference between the antinociceptive effect of each dose of LF and that of saline. Results Intraperitoneally administered LF produced dose - dependent increase of the paw withdraw latency. The peak effect of LF occurred at 60min after the drug administration. The MPE % of all LF groups showed significant differences, compared with that of saline group( P < 0.01). Conclusion Intraperitoneally administered LF has analgesic effect on rat CCI model.
