大豆中异黄酮提取条件及其在提取液中的存在形式

【摘要】 以大豆豆粕为原料,以总异黄酮为目标产物,采用正交试验设计探讨了用乙醇-水体系作溶剂提取大豆异黄酮的最佳条件,即体积分数为70%的乙醇,m乙醇∶V水=0.1g/ml,50℃下提取3次,每次回流时间1 0h.采用酸水解法和等电点沉淀法研究了异黄酮在提取液中的存在形式,即水不溶性异黄酮占8 0%,水溶性异黄酮占92 0%,其中与糖结合的占61 3%,与蛋白质结合的占30 7%,为进一步分离异黄酮提供了试验基础.采用HPLC法测定异黄酮的量.

【Abstract】 The extraction conditions of soybean isoflavones from soybean meal with ethanol-water system are studied. The optimum extracting conditions are obtained with orthogonal experimental design method which are solvent 70% ethanol, the ratio of residue mass to solvent volume 0.1 g/ml, temperature 50 ℃, refluent time 1.0 h and extracting 3 times. The existing forms of isoflavones in extracting solvent are researched with acid hydrolysis and isoelectric precipitation. The results show that the unsolvable isoflavones account for 8.0%; sovable isoflavones 92.0%, the isoflavones combined with sugar and protein account for 61.3% and 30.7% respectively. The concentration of isoflavones is determined by HPLC.
