胞质分裂阻滞微核法对AT细胞高辐射敏感性的研究
Study on high radiosensitivity of AT cells with CBMN method
【摘要】 目的 研究源于毛细血管扩张性共济失调症 (ataxia telangiectasia ,AT)患者皮肤的成纤维细胞系AT5BIVA(AT细胞 )的辐射敏感性。方法 本实验以源于正常人皮肤的成纤维细胞系GM0 63 9(GM细胞 )为对照 ,采用胞质分裂阻滞微核法 (Cytokinesis Blockmicronucleusmethod) ,在AT细胞和GM细胞经60 Coγ射线 0、1、2、3、4Gy照射后 ,观察比较AT细胞和GM细胞之间微核率及微核细胞率的差异 ,并分别进行曲线拟合。结果 在 1、2、3、4Gy剂量照射下 ,AT细胞微核率及微核细胞率均明显高于GM细胞 ,其差异有非常显著性 (P <0 0 1) ,并且两者微核率及微核细胞率均与剂量呈正相关 ,均可拟合成剂量效应直线方程y =a +bx ,微核率及微核细胞率直线回归方程斜率AT细胞明显大于GM细胞 (P <0 0 1)。结论 辐射敏感性AT细胞显著高于GM细胞 ,AT患者AT细胞具有高辐射敏感性。
【Abstract】 Objective To study the radiosensitivity of fibroblast cells (AT cells) from the skin of the patients with ataxia telangiectasia(AT). Methods With Cytokinesis Block micronucleus(CBMN) method, micronucleus frequencies(MNF) and micronucleus cell frequencies(MNCF) of AT cells and GM cells after exposing to 0,1,2,3 and 4 Gy 60 Co γ ray were observed, compared with GMO639(GM cells) from the skin of the normal subjects. The difference of MNF and MNCF between AT cells and GM cells were analyzed, and dose response curves were established. Results The radiation induced levels of MNF and MNCF were significantly higher in the exposed AT cells compared with the GM cells( P <0 01). In the two cell types, both MNF and MNCF had a positive correlation with the doses, and their linear regression equations were: y=a+bx . The slopes of AT MNF and MNCF linear regression equations were larger than those of GM( P <0 01). Conclusion AT cells was extremely sensitive to radiation. The radiosensitivity of AT cells was significantly higher than GM cells.
