双乙烯酮对肝脏和肺脏毒性作用的实验研究

【摘要】 目的 探讨双乙烯酮对肝脏和肺脏的毒作用机制。方法 对大鼠进行吸入染毒 6周 ,然后测定血中丙二醛(MDA)、丙氨酸转氨酶 (ALT)、还原性谷胱甘肽 (GSH) ,以及对大鼠肺支气管灌洗液 (BALF)成分进行测定 ,并对肝脏和肺脏组织进行病理学观察。结果 双乙烯酮染毒大鼠血中MDA含量和ALT活力明显升高 ,GSH含量明显降低。BALF中肺巨噬细胞 (PAM)总数、PAM死亡率、RBC总数、蛋白含量、MDA含量、乳酸脱氢酶 (LDH)活力明显升高 ,磷脂含量明显降低 (P <0 0 5 )。肝脏和肺脏组织也见明显的病理学改变。结论 双乙烯酮具有明显的肝脏和肺脏毒性。

【Abstract】 Objective To explore the toxic mechanism of diketene on liver and lung. Methods Rats inhaled diketene for 6 weeks. Contents of malondialdehyde(MDA), reduced glutathione(GSH) and activity of alanine aminotransferase(ALT) in serum, components in bronchial and alveolar lavage fluids(BALF) were measured and the pathomorphological changes in liver and lung tissues were observed. Results Contents of MDA, activity of ALT increased and contents of GSH decreased significantly in serum. Totality and mortality of pulmonary alveolar macrophage(PAM), RBC, contents of protein, MDA, activity of lactic dehydrogenase(LDH) increased and contents of phosphotide decreased significantly in BALF of rats exposed to diketene(P<0.05). Obvious pathomorphological changes were also observed in livers and lungs. Conclusion Diketene has obvious toxic effects on liver and lung.
