人骨髓间充质干细胞与脐血单个核细胞对混合淋巴细胞反应及PHA诱导转化的抑制

【摘要】 为研究和比较人骨髓间充质干细胞 (MSC)和脐血单个核细胞在体外的免疫调控能力 ,从人骨髓分离培养间充质干细胞 ,用Ficoll分离得到脐血单个核细胞 ,分别以不同剂量加入到外周血混合淋巴细胞培养体系和植物血凝素 (PHA)刺激的外周血淋巴细胞转化体系中 ,用MTT还原法测定细胞增殖 ,分别观察MSC和脐血单个核细胞对混合淋巴细胞反应和淋巴细胞转化的影响。结果显示 ,5× 10 4 ,1× 10 4 MSC以及 2× 10 5脐血单个核细胞均抑制混合淋巴细胞反应和PHA诱导的淋巴细胞增殖 ,而 <1× 10 4 MSC和 <2× 10 5脐血单个核细胞对混合淋巴细胞反应和PHA诱导的淋巴细胞增殖的抑制作用均不明显 ;5× 10 4 MSC对淋巴细胞增殖的抑制能力明显强于 2× 10 5脐血单个核细胞。结论 :大剂量的骨髓间充质干细胞和脐血单个核细胞在体外对混合淋巴细胞反应和PHA诱导的淋巴细胞转化均具有抑制作用 ,而且间充质干细胞的抑制能力强于脐血单个核细胞。

【Abstract】 To study and compare the immunomodulatory functions of human bone marrow mesenchymal stem cell (MSC) and cord blood mononuclear cell (CBMNC) in vitro,human bone marrow MSC were separated with Percoll (1 073 g/L) and cultured in low-glucose DMEM,and cord blood mononuclear cells were isolated with Ficoll (1.077 g/L). These two kinds of cells were added to mixed lymphocyte cultures and PHA induction transformation cultures with various concentrations. The proliferation of lymophocytes was measured by MTT method,effects of MSC and CBMNC on mixed lymphcyte response and PHA-induced lymphocyte transfomation were investigated. The results showed that both 5×104,1×104 MSC and 2×105 CBMNC could inhibit the mixed lymphocyte response (MLR) and PHA induced transformation. But with lower concentrations (MSC≤1×103,CBMNC≤1×105),the inhibition effects of MSC and CBMNC were less consistent. 1×102 MSC and 1×104 CBMNC mainly increased the lymphocyte activation. In addition,the inhibition ratio of 5×104 MSC (MLC 65.3%,PHA 79.1%) was higher than that of 2×105 CBMNC (MLC 8 6%,PHA 37.3%). It is concluded that larger numbers of both MSC and CBMNC showed negative immunomodulatory functions and inhibited the mixed lymphocyte response and induction of transformation in vitro. Moreover,the inhibitory effect of MSC was much stronger than that of CBMNC.
