白矾、氢氧化铝和氯化铝对小鼠学习、记忆及肝肾功能影响的比较研究

【摘要】 目的 比较白矾、氢氧化铝和氯化铝对小鼠学习、记忆和肝肾功能的影响及停药后的恢复情况。方法 白矾[KAl(SO4)2·12H2O]、氢氧化铝[Al(OH)3]和氯化铝(AlCl3)按照铝含量相同的两个剂量14.25 mg/(kg·d)和57 mg/(kg·d),小鼠ig给药60 d后,用高效毛细管电泳仪测定动物血清和脑组织中的铝含量;避暗法测定小鼠学习、记忆能力;分光光度法测定小鼠血清中谷丙转氨酶和尿素氮含量。结果 除白矾小剂量组外,各组小鼠均出现学习、记忆障碍,避暗潜伏期(STL)比对照组明显缩短(P<0.01),血清铝含量显著升高(P<0.01),各大剂量组脑铝含量显著升高(P<0.01),伴血清丙氨酸氨基转移酶(ALT)、尿素氮(BUN)含量显著升高(P<0.01,P<0.05)。停药2周后,学习、记忆障碍仍然存在,STL比对照组明显缩短(P<0.01),但有恢复的趋势;血清铝含量均恢复正常,除氯化铝大剂量组血清BUN及脑铝含量仍高于对照组外(P<0.05,P<0.01),其余各组各指标均恢复正常。结论 白矾、氢氧化铝和氯化铝长期、大剂量给药均可致小鼠学习、记忆障碍,肝、肾功能损伤;氯化铝大剂量组脑铝含量及血清BUN水平恢复较慢,其余各指标均可恢复。

【Abstract】 Objective To study the effects and recovery of alum, aluminum chloride (AlCl3), and aluminum hydroxide [Al(OH)3] on memory, liver and renal function of mice. Methods 60 days after the mice were given daily alum, aluminum chloride and aluminum hydroxide with the same aluminum content of 14.25 mg/(kg·d) and 57 mg/(kg·d), respectively. Aluminum contents in serum and brain of mice were assayed with high performance capillary electrophoresis. The step through latency (STL) was determined with a passive avoidance task. Alanine aminotransferase (ALT) and urea nitrogen (BUN) in serum were assayed with chromatography. Results Except alum small dose group, STL decreased and serum aluminum increased obviously. Brain aluminum, ALT and BUN also increased in all the large dose groups. 2 weeks after the mice were stopped ig these drugs, memory deficits still exist, but have the trend of recovery. Other parameters were recovered to normal level except serum BUN and brain aluminum in aluminum chloride large doses group. Conclusion The intake of alum, aluminum chloride and aluminum hydroxide with large doses (57 mg/kg aluminum) for 60 days induced memory deficits, live and renal function. These deficits could be recovered after the adiministration were stopped, except brain aluminum and serum BUN in aluminum chloride large doses group.
