用作人体植入物的含铈钛合金的生物安全性评价

【摘要】 采用体内急性全身性毒性实验、溶血实验、MTT实验(Tetrazolium basedcolorimetricassay)和材料浸渍液培养细胞的形态学观察法实验对含铈钛合金的生物安全性进行了初步的评价。用X射线光电子能谱仪XPS分析了合金表面氧化膜成分,并用原子发射光谱仪ICP检测了浸渍液中合金元素的种类和浓度。结果显示:含铈和不含铈的Ti Fe Mo Mn Nb Zr系合金表面均形成了以TiO2为主的致密氧化膜,浸渍液中均存在浓度为0.2~0.27mg·L-1的Fe和0.16~0.87mg·L-1的Mn,这种义齿材料有利于补充人体所需的Fe和Mn;两者均未见任何急性毒性反应;溶血程度为0.558%~0.67%,有良好的血液相容性;细胞毒性实验评价级别为0和1级,无明显的细胞毒性作用;倒置相差显微镜观察细胞形态,未发现异常。由以上结果可以初步认为在实验浓度范围内的铈生物安全性是能够保证的。

【Abstract】 The general toxicity tests including acute toxicity test, haemolysis test, MTT assay of Ti-Fe-Mo-Mn-Nb-Zr alloys with Ce and without Ce were investigated. Morphology of these cells was also observed with phase-contrast microscope. Using XPS, the kind and molar ratio of element in surface film were studied. The kind and concentration of element in dipping fluid were investigated by ICP. The results show that the primary component is TiO2 in surface film of both with and without cerium. In the dipping fluid of Ti-Fe-Mo-Mn-Nb-Zr alloys without cerium includes 0.2 mg·L-1Fe and 0.16 mg·L-1Mn, while the dipping fluid of Ti-Fe-Mo-Mn-Nb-Zr alloys with cerium includes 0.27 mg·L-1Fe and 0.87 mg·L-1Mn, so low concentration is beneficial to health. No cytotoxity and toxicity effects were observed in vitro tests. The level of cytotoxicity is grade 0 and 1, and the haemolysis degree of with and without cerium is 0.558% and 0.67%, and both of them are less than 5%. The cells growing in the extract shows normal morphology.
