温度胁迫对甘蓝蛋白质磷酸化的影响
EFFECTS OF TEMPERATURE STRESS ON PROTEIN PHOPHORYLATION IN BRASSICA OLERACEA L.
【摘要】 用SDS-PAGE放射自显影技术以及液闪测定技术研究了2种温度胁迫条件下甘蓝体外蛋白激酶活性变化及其对Ca2+的依赖情况,并进行了温度胁迫磷酸化蛋白的分离。结果表明:甘蓝受到高温(35℃)或低温(5℃)胁迫时,体内蛋白激酶活性在3min内迅速升高,随着温度胁迫时间的进一步延长而迅速降低,发现温度胁迫下,蛋白激酶对Ca2+具有明显的依赖性。从放射自显影结果来看,2种温度胁迫都会发生分子量为22.9kD的特异蛋白质体外磷酸化;在逆境温度处理10min内,磷酸化蛋白质含量较高,随着低温处理时间的延长,磷酸化蛋白质含量迅速下降。
【Abstract】 Protein phosphorylation in vitro of Brassica oleracea L. under two temperature stresses was studied using SDS-PAGR radioautography. The dynamic changes in the activity of protein kinase showed that protein kinase activity reached its maximum within 3 minutes of temperature stress (35℃ or 5℃) and then, with the prolonging of temperature stress, the activity dropped rapidly. Protein kinase activity in situ appeared to be Ca2+-dependent. The extract of B. oleracea plants exposed to 35℃ or 5℃ for 5~10 min showed a 22.9 kD phosphorylation protein band.
