【摘要】 目的　研究卡介苗 (BCG)胞壁蛋白的活性组分能否诱导人单核吞噬细胞 IL - 12 P4 0和 i NOS基因表达。方法　应用半定量逆转录聚合酶链反应 (RT- PCR)法检测 THP- 1细胞 IL - 12 P4 0和 i NOS m RNA在 BCG胞壁蛋白组分刺激下的表达变化。结果　卡介苗胞壁蛋白 Sephadex G- 15 0层析分子量约 2 1～ 2 9k D的组份可诱导 THP- 1细胞 IL -12 P4 0和 i NOSm RNA的表达增强。结论　 BCG胞壁蛋白可刺激人单核吞噬细胞 IL - 12 P4 0和 i NOS基因的增强表达。更多还原

【Abstract】 Objective To examine the role of BCG cell wall proteins in the induction of IL 12p40 and iNOS mRNA expression in human monocytes. Methods Using semi quantitative reverse transcription PCR (RT PCR), the induction of IL 12p40 and iNOS mRNA expression was detected in THP 1 cells stimulated with Sephadex G 150 fractions of BCG cell wall protein. Results A markedly increase in IL 12p40 and iNOS mRNA expression was detected by semi quantitative RT PCR in THP 1 cells when stimulated with a fraction (a molecular range of 21 29 kD ) of BCG cell wall proteins. The inducedexpression of IL 12 p40 and iNOS mRNA was dose and time dependent. Conclusion This experiment provides evidence that BCG cell wall proteins can stimulate IL 12p40 and iNOS mRNA expression in human monocytes.更多还原
