CAMKⅡD基因在3T3-L1脂肪细胞诱导分化过程中的表达变化
Changes of Calcium/Calmodulin-Dependent Protein Kinase Ⅱδ Gene Expression during Differentiation of 3T3-L1 Preadipocyte
【摘要】 目的 观察 3T3 L1前体脂肪细胞诱导分化过程中Ⅱ型钙调蛋白激酶δ亚基 (CAMKⅡD)基因表达水平的变化 ,探讨CAMKⅡD基因与肥胖发生的关系。方法 体外培养 3T3 L1前体脂肪细胞 ,在诱导 3T3 L1脂肪细胞分化成熟的不同时段 ,采用RT PCR技术检测脂肪细胞中CAMKⅡD基因的mRNA表达水平。结果 诱导剂刺激后 (第 0天 ) ,3T3 L1脂肪细胞中CAMKⅡD基因的mRNA表达于第 1天显著下调 ,与第 0天比较具有显著差异 (P <0 .0 1) ;第 2天 ,该基因表达水平显著上调 ,并明显高于第 0天表达水平 (P <0 .0 1) ;第 2天至 3T3 L1细胞分化为成熟脂肪细胞 (第 10天 ) ,该基因表达一直维持在较高水平 (P >0 .0 5 )。结论 CAMKⅡD基因参与了脂肪细胞分化的调控 ,与肥胖发生相关 ,其在3T3 L1细胞分化过程中的表达变化可能有利于脂肪细胞的分化成熟和脂质积聚
【Abstract】 Objective To investigate the changes of calcium/calmodulin-dependent protein kinase Ⅱδ (CAMKIID) gene expression during differentiation of 3T3-L1 preadipocyte and to explore the relationship between CAMKⅡD gene and the occurence of obesity.Method 3T3-L1 preadipocytes were cultured in vitro and differentiated into the matured adipocytes.Total RNA was extracted from adipocytes at various stages and the expression level of CAMKⅡD gene mRNA was evaluated by RT-PCR.Results Under the stimulation of inducing reagents(Day 0),the level of CAMKⅡD gene mRNA expression in 3T3-L1 adipocytes was significantly down-regulated at Day 1,when compared with that at Day 0(P<0.01).However,the marked up-regulation of CAMKⅡD gene mRNA expression in 3T3-L1 adipocytes was observed at Day 2.Compared with that at Day 0,the level of CAMKⅡD gene mRNA expression was significantly higher at Day 2(P<0.01).Thereafter,the level of CAMKⅡD gene mRNA expression remained high from Day 2 to Day 10(the matured adipocytes,P>0.05).Conclusions CAMKIID gene is involved in the differentiation of adipocytes and related to the occurence of obesity.The changes of CAMKⅡD gene mRNA expression level during differentiation of 3T3-L1 preadipocyte may be helpful to the differentiation and adipogenesis of adipocytes.
