用凝胶电泳研究发夹型引物检测端粒酶活性
Gel electrophoresis studing hairpin structure primer on gene detection of predictive marker for malignant tumor-telomerase activity
【摘要】 端粒酶是目前最广谱的恶性肿瘤的预警标志物 ,其活性对大多数恶性肿瘤的诊断均具有较高的敏感性和特异性。本文针对端粒酶延伸产物序列的特殊性 ,开发一类新型发夹型引物。该引物具有的发夹结构使得PCR扩增只能在加热条件下启动。运用这项技术 ,建立用非变性聚丙烯酰胺凝胶电泳检测端粒酶活性的新方法。减少非特异性扩增产物及引物二聚体的生成 ,提高检测的专一性。将该法用于恶性肿瘤细胞提取物中端粒酶活性的检测 ,取得满意的结果。
【Abstract】 Telomerase is the most universal predictive marker for human cancer.Telomerase activity has high sensitivity and specificity for diagnosis of most malignant tumors.Aiming at the specific sequence of telomersae product,a novel primer with hairpin structure has been designed and synthesized.It denatures to a linear structure and serves as an effective primer only at high temperature.Using this technology a novel method for detection of telomerase activity has been established using nondenaturing gel electrophoresis.It enhances the specificity of detection due to decreasing the products from non-Specific amplification and primer dimerization.The method has been used to detect telomerase activity in the extracts of malignant tumor cell with satisfactory results.
