小檗碱对小鼠耐缺氧作用的影响
Anti-hypoxia activity of berberine in mice
【摘要】 目的:研究小檗碱对小鼠耐缺氧作用的影响及其机制。方法:①小鼠(n=40)随机分为4组,分别腹腔注射小檗碱0.1mg/g,0.2mg/g,15min后,将小鼠分别放入250mL的磨口广口瓶内密封,以最后一次呼吸为指标,观察小鼠存活时间。②小鼠(n=30)随机分为3组,分别腹腔注射小檗碱0.1mg/g,0.2mg/g,15min后,不需麻醉,用剪刀在耳下部快速断头,记录喘气时间。③小鼠(n=30)随机分为3组,对照组按0.03mL/g剂量给小鼠皮下注射生理盐水溶液,5min后按0.02mL/g剂量给小鼠腹腔注射生理盐水;模型组按0.015mg/g的剂量给小鼠皮下注射异丙肾上腺素,5min后按0.02mL/g剂量给小鼠腹腔注射生理盐水溶液。给药组按0.015mg/g剂量给小鼠皮下注射异丙肾上腺素,5min后腹腔注射小檗碱(0.1mg/g)。给药后10min,分别放入广口瓶内密封,记录小鼠存活时间。结果:小檗碱(0.1,0.2mg/g)能非常显著延长常压耐缺氧条件下小鼠的存活时间犤(67.8±11.2)和(81.2±13.5)min犦,与对照组犤(41.3±5.2)min犦比较,差异均有显著性意义(t=3.112,3.312;P均<0.01);小檗碱(0.1mg/g,0.2mg/g)能显著延长断头小鼠的呼吸时间犤(22.8±1.6)和(26.1±3.1)s犦,与对照组犤(18.5±1.6)s犦比较,差异有显著性意义(t=2.654,3.423;P<0.05~0.01);小檗碱(0.1mg/g)能非常显著延长皮下注射异丙肾
【Abstract】 AIM: To study the anti hypoxia effect of berberine and its mechanism in mice.<METHODS:① Forty mice were randomized into four groups, and berberine at the doses of 0.1 mg/g and 0.2 mg/g was injected intraperitoneally respectively.After 15 minutes, the mice were put into jars and sealed.The last breath was used as index to observe the survival time of mice.② Thirty mice were randomized into three groups,and given intraperitoneal injection of berberine(0.1 mg/g and 0.2 mg/g).After 15 minutes, the heads of mice without anesthesia were cut quickly at infraauricular region.The breath time was recorded.③ Thirty mice were randomized into three groups:the control group received subcutaneous injection of normal saline(0.03 mL/g) 5 minutes before intraperitoneal injection of normal saline(0.02 mL/g);the model group received Subcutaneous injection of isoproterenol(IP,0.015 mg/g),and then intraperitoneal injection of normal saline(0.02 mL/g) after 5 minutes; the medication group received subcutaneous injection of IP(0.015 mg/g),and then intraperitoneal injection of berberine after 5 minutes.Ten minutes after administration,the mice were taken into the jars and sealed.The survival time was recorded.<RESULTS:Berberine( 0.1 and 0.2 mg/g) could obviously pro1ong the survival time of mice under the normal pressure and hypoxia condition[(67.8± 11.2) and ( 81.2± 13.5) minutes respectively) ,significantly longer than that of the control group[(41.3± 5.2) minutes](t=3.112,3.312, P< 0.01).Berberine( 0.1 mg/g and 0.2 mg/g) could also obviously pro1ong the breath time of mice after decapitation as compared with the control group[(22.8± 1.6) and ( 26.1± 3.1) s vs( 18.5± 1.6) s](t=2.654,3.423, P< 0.05- 0.01).Berberine( 0.1 mg/g) could also prolong the survival time of mice after the subcutaneous injection of IP under the common pressure and hypoxia,and there was a significant difference from the control group [(41.1± 2.1) vs ( 37.1± 1.8) minutes](t=2.663, P < 0.05) .<CONCLUSION:Berberine plays a remarkable role in anti hypoxia.
