大鼠脑出血模型中神经细胞凋亡的特征及Bcl-2蛋白对细胞凋亡的调控
Characteristics of nerve cell apoptosis and effect of Bcl-2 protein expression on the regulation and control of apoptosis in rats with intracerebral hemorrhage
【摘要】 目的:研究大鼠脑出血灶周围是否存在细胞凋亡及Bcl-2蛋白的表达。方法:SD大鼠50只,随机分为5组,10只为对照组,其余40只建立大鼠脑出血模型,分别于模型制备后12,24,48,72h取脑组织,应用末端脱氧核苷酸转移酶介导的脱氧三磷酸尿苷缺口末端标记法(TUNEL)检测细胞凋亡,免疫组化SP法检测Bcl-2蛋白。结果:正常细胞对照中未见TUNEL及Bcl-2阳性细胞,其余各组中TUNEL及Bcl-2阳性细胞均有不同程度表达。凋亡率随出血后时间不同而发生相应改变,在出血后48~72h达到高峰,TUNEL阳性细胞率峰值为(24.50±2.69)%,实验组与对照组间差异有显著性意义(P<0.05)。Bcl-2蛋白在出血灶周围细胞中亦有表达,其阳性表达率峰值为(20.76±1.97)%,且Bcl-2蛋白的表达与凋亡细胞之间并无明显线性关系。结论:细胞凋亡参与了脑出血后神经细胞的继发损伤过程,其在出血后的表达存在一个时间窗,对临床及时治疗有一定指导意义;Bcl-2蛋白在出血后一定时间内持续升高,但与细胞凋亡不同步,表明了bcl-2的抑制细胞凋亡的作用。
【Abstract】 AIM:To study whether there is apoptosis and expression of Bcl-2 protein or not in rats with intracerebral hemorrhage(ICH). METHODS:Fifty SD rats were divided into 5 groups.Ten were in the control group,the other 40 rats were made into ICH models,and the brain was taken 12,24,48 and 72 hours after the establishment of models,then the apoptosis was detected by terminal deoxynucleotidyl transferase mediated dUTP nick end labeling(TUNEL),and the expression of Bcl-2 protein was detected by immunochemical staining. RESULTS:No TUNEL and Bcl-2 positive cells were observed in the normal control group,while there were TUNEL and Bcl-2 proteins of different levels in other groups.The apoptosis rate varied correspondently with the increase of time after ICH,and reach its peak 48 to 72 hours after ICH,the peak positive rate of TUNEL cells was(24.50±2.69)%,and there was significant difference between the experimental groups and control group(P< 0.05).Bcl-2 protein was expressed in the neural cells near the hemorrhage,and the peak positive rate of the Bcl-2 protein expression was(20.76±1.97)%.There was no obvious linear relation between apoptosis and the expression of Bcl-2 protein. CONCLUSION:Apoptosis participates in the secondary trauma of nerve cells after ICH,and there is a time window of its expression after ICH,which has certain instructive significance to the clinical therapy in time.The expression of Bcl-2 protein keeps increasing during certain period after ICH but not synchronous with apoptosis,indicating that bcl-2 has the effect in inhibiting apoptosis.
