硒和维生素E对实验性高脂血症大鼠体内部分抗过氧化物酶的影响
Effects of selenium and vitamin E on some antioxidant enzymes in experimental hyperlipidemic rats
【摘要】 目的:探索硒和(或)维生素E对高脂饲料所致实验性高脂血症大鼠心、肝、肾及血清超氧化物歧化酶(superoxidedismutase,SOD)、谷胱苷肽过氧化酶(glutathioneperoxidase,GSH-Px)、过氧化氢酶(catlase,CAT)等抗过氧化酶的影响作用,探讨硒和维生素E抗脂质过氧化的作用。方法:采用高脂饲料致大鼠实验性高脂血症,然后分别分组给予经预试后的硒和/或维生素E,4周后行乙醚麻醉取血,同时取心、肝、肾组织作匀浆。采用测试试剂盒对血清、心、肝及肾组织匀浆进行SOD、GSH-Px、CAT测定。结果:高脂饲料可不同程度地降低血清、心、肝及肾组织的SOD、GSH-Px、CAT活力,与常规饲料组相比,肝脏的GSH-Px(P<0.05),血清CAT(P<0.01)有统计学差异;而单独或联合使用硒和维生素E可对抗高脂饲料所致的这一作用,明显增强这些抗氧化酶的活力,与高脂饲料组相比有显著性差异(P<0.05或P<0.01),且二者合用的作用明显强于各自单独使用,如肝脏及血清GSH-Px(P<0.05或P<0.01);心、肾脏及血清CAT(P<0.05,P<0.01或P<0.001);肾脏SOD(P<0.01)。结论:硒和维生素E单独或联合应用可对抗或增强实验性高脂血症大鼠SOD、GSH-Px、CAT活力。
【Abstract】 AIM:To study the effects of selenium(Se) and/or vitamin E(VE) on heart, liver,kidney and antioxidant enzymes,such as superoxide dismutase(SOD),glutathione peroxidase(GSH Px) and catlase(CAT) in experimental hyperlipidemic rats,and the effects on anti peroxidation of grease. METHODS:SD rats were used to make experimental hyperlipidemia models with hyperlipidemic feed,and then divided separately.Rats were given Se and/or VE in different groups after prediction.Four weeks later, rats were anesthetized with ether,and their blood was extracted with homogenate in hearts,livers and kidney,and SOD,GSH Px and CAT were assayed. RESULTS:SOD,GSH Px and CAT activities were differently reduced in all the experimental tissues by hyperlipidemic feed,such as serum, heart,liver and kidney,and GSH Px in liver(P< 0.05) and CAT in serum(P< 0.01) were significantly different from those in the normal feed group.However,Se and/or VE could fight against the role of hyperlipidemic feed and increase the activity of antioxidant enzymes,which was significantly different compared with hyperlipidemic feed group(P< 0.05 or P< 0.01).Combined usage of Se and VE was more effective, such as GSH Px in livers and serums(P< 0.05 or P< 0.01);CAT in heart, kidney and serum(P< 0.05,P< 0.01 or P< 0.001);SOD in kidney(P< 0.01). CONCLUSION: Se and/or VE can fight against or increase the SOD, GSH Px and CAT activities in experimental hyperlipidemic rats.
