脑心通胶囊对冷水负重游泳模型小鼠的心、脑保护作用

【摘要】 目的:观察脑心通胶囊对冷水负重小鼠冷水中存活时间的影响,探讨其对模型鼠脑及心脏保护作用的机制。方法:①实验动物按分层随机法分为5组,即模型对照组、天保宁片40mg/kg组、脑心通胶囊0.4g/kg组、0.8g/kg组、1.6g/kg组,小鼠8℃冷水负重游泳至死,测定存活时间。②实验动物按分层随机法分为6组,即正常对照组、模型对照组、天保宁片40mg/kg组、脑心通胶囊0.32g/kg组、0.64g/kg组、1.28g/kg组,小鼠8℃冷水负重游泳5min,测定脑组织及心脏组织生化代谢的改变。结果:①脑心通胶囊0.8和1.6g/kg可以明显延长模型小鼠的存活时间,分别是(845.00±122.23),(749.38±131.04)s,与模型对照组(620.38±75.35)s比较,差异有非常显著性意义(t=-6.204,-3.414,P<0.01)。②与正常对照组相比,模型小鼠脑组织乳酸含量增加,Na+-K+-ATP酶活力降低,一氧化氮合酶(NOS)活力增加(P<0.05),一氧化氮含量未见明显变化;心脏组织乳酸含量增加,Na+-K+-ATP酶活力和肌酸激酶活力降低(P<0.05)。脑心通胶囊可以不同程度地逆转模型小鼠脑组织及心脏组织上述生化指标的异常改变(P<0.01)结论:脑心通胶囊可延长冷水负重游泳模型动物的存活时间,改善脑组织及心脏组织生化代谢可能是其发挥脑保护作用和心脏保护作用的部分机制。

【Abstract】 AIM:To observe the effect of naoxintong capsule on the survival time of loaded mice in cold water,so as to explore its mechanism for protecting brain and hear t. METHODS:① In order to determine the survival time,heavy-loaded mice were ma de to swim in cold water(8 ℃) until death, and the mice were divided into 5 gro ups with the stratified random sampling: model control group,tianbaoning tablet group(40 mg/kg),naoxintong capsule 0.4 g/kg,0.8 g/kg and 1.6 g/kg groups,then th e survival time was recorded.②Mice were divided into 6 groups with the stratifi ed random sampling: normal control group,model control group, tianbaoning tablet group(40 mg/kg),naoxintong capsule 0.32 g/kg,0.64 g/kg and 1.28 g/kg groups.Aft er heavy-loaded swimming for 5 minutes, the changes of biochemical metabolism i n brain and heart tissues were assayed. RESULTS:①The survival times in the naoxintong capsule 0.4 g/kg,0.8 g/kg group s were obviously prolonged[(845.00±122.23),(749.38±131.04) s],significantly di fferent from that in the model control group[(620.38±75.35) s](t=6.204,-3.414, P< 0.01).②Compared with the normal control group,the content of lactic acid (LA ) in brain tissue increased, the activity of Na+-K+-ATPase decreased,while the activity of nitricoxide synthase (NOS) increased(P< 0.05),but the content of nitrogen monoxide(NO) did not change in brain tissue of the model control group ; In the heart tissue of model mice,the content of LA increased, the activities of Na+-K+-ATPase and creatine kinase decreased(P< 0.05).Naoxintong capsule c ould reverse the above abnormal biochemical indexes in brain and heart tissues t o different extent(P< 0.01). CONCLUSION:Naoxintong capsule can increase the survival time of heavy-loaded model mice swimming in cold water,and improve the biochemical metabolism of brai and heart,which may be the partial mechanism of its protective effect on brain and heart.
