呆聪液对老龄痴呆动物模型影响的研究

【摘要】 目的:探讨呆聪液对老龄痴呆大鼠动物模型脑M受体、某些血清酶活性与过氧化脂质(lipidperoxides,LPO)及免疫功能的影响。方法:采用双侧电解损毁22月龄大鼠脑基底核法,造成动物学习与记忆功能障碍的老龄鼠痴呆动物模型,用中药呆聪液灌胃给药1个月后,处死动物测定脑组织M受体;检测血清中超氧化物歧化酶(superoxidedismutase,SOD)、胆碱酯酶和单胺氧化酶(monoamineoxidase,MAO)的活性水平及LPO的含量;用3H-TdR掺入DNA法测定淋巴细胞增殖能力。结果:呆聪液可提高老龄痴呆大鼠脑M受体数目(P<0.01);提高血清SOD活性:呆聪液10g/(kg·d)组,SOD活性为(173.02±23.07)NU/mL,与模型组(75.75±11.21)NU/mL比较,差异有非常显著性意义(q=15.13,P<0.01)。呆聪液可降低LPO含量(P<0.01),降低MAO和胆碱酯酶活性(P<0.01);提高淋巴细胞增殖能力(P<0.01)。结论:呆聪液明显提高老龄痴呆大鼠脑M受体数目,对氧自由基具有一定的清除作用,还可清除LPO和抑制胆碱酯酶与MAO的活性,具有益智抗衰老及提高免疫功能的作用。对老年性痴呆有较好的疗效。

【Abstract】 AIM:To explore the effect of daicong sloution on brain M receptor, some serum enzyme activity,lipid peroxides(LPO) and the immunity of aged dementia rat models. METHODS:22 month old rats whose basal nuclei were destroyed by bilateral electrolytic method were used as aged dementia rat models which had learning and memory disability.They were treated with daicong solution for a month.Then the rats were killed and the number of M receptors in the brain tissue and the activities of superoxide dismutase(SOD),cholinesterase(CHE) and monoamine oxidase(MAO),and LPO level in the serum were measured. CONCLUSION:Daicong solution can obviously increase the number of M receptors in the brain tissue of aged dementia rats and has some certain clear function to oxygen derived free radidicals.Furthermore,it can clear LPO and inhibit the activity of CHE and MAO,and thus helps to enhance intelligence and slow aging and increase immunological function.It is a comparatively effective compound in treating aged dementia.
