电针对脑缺血大鼠血清白细胞介素-6的影响及其意义
Effect of electroacupuncture on serum interleukin-6 in rats with cerebral ischemia and its significance
【摘要】 目的:观察脑缺血后不同时间段白细胞介素-6(Interleukin-6,IL-6)在血清的表达及电针对其表达的调节作用。方法:采用随机数字表法将大鼠分为10组正常对照组,假手术组,缺血1,3,7,14d4组,缺血1,3,5,7,14d+电针4组,每组8只,通过大脑中动脉线栓法建立SD大鼠局灶性脑缺血模型,于造模成功后电针缺血+电针组大鼠的足三里、内关,再利用放射免疫分析技术,检测脑缺血及电针对IL-6表达的影响。结果:放射免疫分析显示,在正常和假手术组大鼠血清IL-6呈基础水平表达。缺血后IL-6表达迅速增加,1d达高峰,缺血1d,3d组和正常对照组比较差异有非常显著性意义(t=4.828,2.984,P<0.01)。脑缺血后3,7d,缺血组和相应时间点缺血+电针组比较,差异有显著性意义(t=2.622,2.484,P<0.05)。结论:电针提高血清IL-6的表达可能是电针治疗缺血性脑损伤的机制之一。
【Abstract】 AIM:To investigate the expression of serum interleukin 6(IL 6 )and the effect of electroacupuncture(EA) on IL 6 expression at various time points in rats after cerebral ischemia. METHODS:SD rats were divided into ten groups(n=8) with simple random sampling method: normal control group, sham operation group,ischemia 1,3,7,14 days groups, ischemia+EA groups after ischemia 1,3,7,14 days.The middle cerebral artery of SD rat was occluded by a monofilament to prepare focal cerebral ischemia model. After that,EA was applied at zusanli and neiguan of rats in ischemia+EA groups.Then the influence of cerebral ischemia and EA on IL 6 expression of rats was detected with radioimmunoassay method. RESULTS:Rats in the normal control group and sham operation group showed a basic expression of IL 6 in serum.The concentration of serum IL 6 increased quickly after ischemia and reached the peak 1 day later.There was significant difference between ischemia 1,3 days groups and normal control group(t=4.828,t=2.984,repectively,P< 0.01).At 3,7 days of ischemia,there was a significant difference between ischemia groups and ischemia+EA groups(t=2.622,t=2.484,respectively,P< 0.05). CONCLUSION:The upregulation of IL 6 expression may be one of the mechanisms of EA in treating ischemic cerebral damage.
