性别因素对应激大鼠下丘脑c-fos表达的影响

【摘要】 目的:探讨性别因素对下丘脑室旁核中Fos表达的影响,以期揭示应激相关障碍的中枢机制。方法:雌、雄wistar大鼠各15只随机分为4组:正常对照组10只,分为雌(A)、雄(B)2组各5只;束缚应激实验组20只,分为雌(C)、雄(D)2组各10只,束缚应激组大鼠束缚在特制的限制器内3h,对照组不予束缚处理;用免疫组织化学方法检测应激前后的大鼠室旁核Fos的表达。结果:雌、雄性应激组大鼠下丘脑c-fos基因表达阳性细胞百分率分别为(61.2±2.5)%,(61.4±2.4)%,两组比较差异无显著性意义(t=0.066,P>0.05),不同性别大鼠应激后下丘脑室旁核Fos阳性细胞数量无显著差别。结论:性别因素可能和应激相关障碍无关。

【Abstract】 AIM:To study the effect of gender on the Fos expression in the hypothalamus of rat in order to disclose the central mechanism of immocilization stress related disorder. METHODS:Female and male rats(15 each) were randomly divided into four groups: normal group(n=10) which was divided into two subgroups: group A(5 females), group B(5 males), immobilization group(n=20) which was also divided into two subgroups: group C(10 females)and group D(10 males).Rats in the immobilization group were limited in a special designed killer for 3 hours,while rats in the control group were without bondage.The Fos expression in hypothalamus was detected by immunohistochemical method. RESULTS:The rate of c fos positive cells in the hypothalamus of rat after immobilization stress in group C and group D were(61.2±2.5)%and(61.4±2.4)%,respectively, and there was no significant difference between the two groups(t= 0.066,P >0.05).There was no obvious difference in the number of Fos positive cells in hypothalamus of rats of different gender after immobilization stress. CONCLUSION:Gender may have no correlations with stress related disorder.
