降钙素基因相关肽对全脑缺血脑组织的保护作用

【摘要】 目的:通过降钙素基因相关肽(calcitoningenerelatedpeptide,CGRP)对实验小鼠能量代谢的研究,进一步探讨CGRP对缺血脑组织的保护作用。方法:实验小鼠在缺血前给予CGRP,并在处死后测定缺血脑组织的三磷酸腺苷(ATP)和乳酸含量。ATP的测定采用虫荧光素-荧光素酶生物发光法;乳酸的测定采用对羟基联苯显色法。脑缺血模型的制作:采用小鼠断头缺血法。结果:CGRP对实验小鼠全脑缺血有明显的预防保护作用,表现为:CGRP可使缺血脑组织中乳酸水平减少,使ATP水平明显增加。而这种作用与给药剂量和给药方式有关,即在CGRP的剂量为2.67BU/g时效果较好,ATP为(5.4±2.3)pmol/g,乳酸为(1.4±0.4)mg/g,与模型组(2.6±1.3)pmol/g,(3.2±2.0)mg/g比,差异有显著性意义(P<0.05)。且随着给药时间的延长,缺血脑组织中乳酸水平降低更加明显(P<0.05),ATP水平显著增高(P<0.05)。CGRP对小鼠缺血脑组织的保护作用在超过一定剂量后无显著作用。结论:CGRP对全脑缺血小鼠有确切的预防保护作用,但应掌握合适剂量。

【Abstract】 AIM:To investigate the effect of calcitonin-gene-related peptide(CGRP) in pr otecting brain tissue after cerebral ischemia on the basis of the study on the e ffect of CGRP on energy metabolism of experimental mice. METHODS:Experimental mice were administrated CGRP before cerebral ischemia,and contents of adenosine triphosphate(ATP) and lactic acid in ischemic tissue were determined after decapitation.ATP was assayed by using luciferine-fluoresceina se bioluminescent method,and lactic acid by using p-hydroxy-diphenyl method.Gl obal cerebral ischemic model was made by decapitation. RESULTS:CGRP had an obvious role in preventing and protecting global cerebral ischemia,which could decrease the content of lactic acid and increase the conten t of ATP in brain tissue.The role of CFRP was associated with the dose and way o f administration.When CGRP was at a dose of 2.67 BU/g,the contents of ATP and la ctic acid were(5.4±2.3) pmol/g and(1.4±0.4) mg/g respectively,significantly hi gher than those in the model group[(2.6±1.3) pmol/g and(3.2±2.0) mg/g respecti vely](P< 0.05). The prolonged time of administration of CGRP had a significant e ffect in decreasing the content of lactic acid in brain tissue(P< 0.05),and incr easing the content of ATP(P< 0.05).CGRP over a certain dose had no protective effect on ischemic brain tissue. CONCLUSION:CGRP can protect brain from damage of global cerebral ischemia.Howe ver its dose should be confined in a proper level.
