去睾丸大鼠骨密度和骨组织形态计量法参数的相关性

【摘要】 目的:检测去睾丸大鼠的骨密度及骨组织形态计量法静态参数,并且将各参数行组间对比研究以及骨密度与骨组织形态计量法静态指标相关性研究,探讨检测大鼠骨质疏松的方法。方法:选用19只三四月龄的Wistar雄性大鼠,随机分成去睾组和对照组,同条件下饲养18周。处死前用双能量X线骨密度仪)测量大鼠腰椎、股骨颈、股骨粗隆和股骨近端的骨密度。大鼠处死后取左侧股骨近端行塑料包埋的骨组织切片及组织形态计量法分析。结果:去睾丸大鼠18周后,L4-5骨密度减少33.8%,股骨颈减少29.1%,粗隆部减少29.3%,股骨近端减少30.1%,与对照组比较差异有显著性意义(t=-4.90,-3.35,-3.94,-4.65,P均<0.01)。去睾组的骨小梁面积百分率减少46.4%,骨小梁数目减少43.8%,骨小梁分离度增加144.6%,与对照组比较均有显著性差异(t=-8.42,-8.75,7.49,P均<0.01)。股骨颈的骨小梁面积百分率、骨小梁数目和骨小梁分离度均与各个部位的骨密度有相关性,其中与腰椎的骨密度相关性最强(r=0.61,0.65,-0.69,P均<0.01)。结论:去睾丸大鼠腰椎部位的骨密度与股骨颈的骨组织形态计量法有更好的相关性,大鼠腰椎的骨密度检查有方便、经济、敏感的特点。

【Abstract】 AIM:To discuss the better method of examining osteoporosis in rats,by analyzing bone mineral density(BMD) and bone static histomorphometry of orchidectomized rats,and taking contrastive investigation in every parameters intragroup,and studying the relation between them. METHODS:Nineteen male Wistar rats,aged 3-4 months old were randomly divided into two groups: sham surgery(Sham) and orchidectomy group,and raised 18 weeks at same conditions.BMD of lumbar,neck of femur,femur tuberosity and proximal femur in all rats were determined by Hologic QDR 2000+DEXA before sacrifice.Left proximal femur eliminated soft tissues was embedded in methyl methacnylate and cut into slices,and then bone histomorphometry of femoral neck was analyzed. RESULTS:BMD of L4-5,neck of femur,tuberosity and proximal femur were decreased by 33.8%,29.1%,29.3%and 30.1%respectively,significantly different from those of the control group(t=-4.90,-3.35,-3.94,-4.65,P< 0.01).The static parameters such as percentage of trabecular area,trabecular number of the orchidectomy group decreased by 46.4%and 43.8%,while trabecular separation increased by 144.6%,significantly different from those of the control group(t=-8.42,-8.75,7.49,P< 0.01).The percentage of trabecular area,trabecular number and trabecular separation of the neck of femur were related to the BMD of all parts of the rats,and the BMD of lumber was the best interrelated part (γ=0.61,0.65,-0.69,P< 0.01). CONCLUSION:There is a better correlation between the BMD of lumbar and the bone histomorphometry of the femoral neck in orchidectomized rats.The examination of the BMD of lumbar is convenient,economic and sensitive.
