【摘要】 目的:探索BLCAP基因反义寡核苷酸对人骨肉瘤细胞SOSP-9607的影响,为保肢手术方法作前瞻研究基础。方法:通过细胞培养,测量细胞生长曲线、计算细胞集落形成率、检测细胞周期,观察BLCAP基因反义寡核苷酸对人骨肉瘤细胞系SOSP-9607的影响。结果:BLCAP基因反义寡核苷酸浓度为10μmol/L时,对此细胞的生长速度和集落形成能力有明显促进效应;流式细胞仪检测发现其引起细胞周期变化,对细胞增殖活性有增强作用。结论:首次发现BLCAP基因反义寡核苷酸对人骨肉瘤细胞SOSP-9607有明显的影响,提示BLCAP基因可能作为一种抑癌基因在人骨肉瘤发展过程中起作用。

【Abstract】 AIM:To explore the effect of BLCAP antisense oligonucleotide on human osteosarcoma cell line SOSP 9607 to provide a prospective research rationale for extremity saving operations. METHODS:Using BLCAP antisense oligonucleotide on cultured human osteosarcoma cell line SOSP 9607. The cell growth curve was depicted, cell colony forming rate was calculated, and cell cycle was measured. RESULTS:Cell growth and colony forming ability were apparently stimulated when the content of BLCAP antisense oligonucleotide was 10 μmol/L. Also, flow cytometry indicated that the cell cycle changed and it promoted cell proliferation activity. CONCLUSION: The present study found for the first time that BLCAP antisense oligonucleotide had an obvious effect on human osteosarcoma cell line SOSP 9607, suggesting that the gene BLCAP may function as a anti oncogene in the development of human osteosarcoma.
