中国人肝细胞系作为生物人工肝细胞材料的可行性探讨

【摘要】 目的:探讨国内人肝细胞系作为生物人工肝细胞材料的可行性。方法:收集国内9株人肝细胞系,体外培养,光镜、电镜下观察细胞生长分化特性;以流式细胞仪测定其DNA指数值;测定培养上清中人白蛋白、尿素含量及安定转化量;将CL-1细胞接种到裸鼠皮下,观察其致瘤性。结果:9株人肝细胞系中,CL-1细胞分化最好,DI值与正常人淋巴细胞的DI值最接近,生物合成及代谢功能最好,但有一定的致瘤性。结论:9株人肝细胞系中,CL-1人肝细胞系最可能用作BAL细胞材料,有进一步研究的价值。

【Abstract】 AIM:To investigate t he feasibility of human liver cell lines in China used as cell materials of bioart ificial liv er(BAL).METHODS:Nine human liver cell lines were coll ected(nam ed cell line 1to cell line 9respectively,CL-1to CL-9)and cultured in vit ro,then their growth characteristics observed by light microscope and electron micro scope to judge the differentiation of them.DNA index(DI )were determine d by flow cytometer(FCM).The quantity of human albumin,urea synthesis and di -azepam transformed by the human liver cell lines in the supernate were de-ter mined as a probe of the biological function.The oncogenicity of CL-1w as det ermined by the pathological exam ination of tissue which was extracte d from the subcutaneous nodule of nude mice.RESULTS:CL-1had the characteristics of the DI which was close to the normal human lymphocytes and had good biological func tions of synthesis and metabolism but a little oncogenicity.Among t he 9human liver cell lines,CL-1can possibly be used as the cell material of BAL,whil e further research should be done be-fore it being used clinically.
