孕期被动吸烟对大鼠新生鼠仔海马超微结构及NMDA受体NR1亚基的影响
Effects of prenatal passive smoking on the ultrastructure and NMDA receptor NR1 subunit in the neonatal rat hippocampus
【摘要】 目的 :探讨孕期被动吸烟对大鼠新生鼠仔海马CA1区超微结构及N 甲基 天冬氨酸受体 (NMDA)NR1亚基mRNA表达的影响。方法 :将SD雌性大鼠随机分为实验组和对照组。实验组于妊娠第 5~ 2 0日制造被动吸烟模型。取新生鼠仔海马CA1区组织常规电镜制片 ,观察其超微结构 ;用逆转录聚合酶链反应 (RT PCR)技术半定量检测海马NMDA受体NR1亚基mRNA的表达。结果 :实验组新生鼠仔海马CA1区超微结构的改变有 :(1)神经细胞丧失完整结构 ,胞膜溶解 ;(2 )线粒体肿胀 ,嵴断裂、消失 ;(3)粗面内质网肿胀 ;(4)染色质聚集成大小不等的分散团块状 ,附着在核膜周围。实验组新生鼠仔海马NR1亚基mRNA表达水平为 0 .3811± 0 .10 4 3,低于正常对照组的 0 .9318± 0 .15 93,两者差异有显著性 (P <0 .0 5 )。结论 :孕期被动吸烟可引起新生鼠仔海马CA1区超微结构改变和NMDA受体NR1亚基mRNA表达降低 ,这可能是导致神经功能受损的机制之一。
【Abstract】 Objective:To explore the effects of prenatal passive smoking on the ultrastructure of CA1 regions and the expression of NMDA receptor NR1 subunit mRNA in the neonatal rat hippocampus.Methods:Female Sprague-Dawley rats were randomly divided into the test group(TG) and the control group(CG).TG was subjected to passive smoking from gestation day 5 to 20.The CA1 regions in the neonatal rat hippocampus were prepared for routine electron microscopic section to observe the morphologic details.The expression of NMDA receptor NR1 subunit mRNA was semi-quantitatively determined by reverse transcription-polymerase chain reaction (RT-PCR).Results:The electron microscopic changes in the CA1 regions of TG were as followed:(1)the loss of integrity and dissolution of the cell membrane in neurons;(2)the swelling and disruption and/or loss of cristae in mitochondria;(3)the swelling of rough endoplasmic reticulum;(4)the aggregation of chromatins into different sized discrete masses,attaching around the nuclear membrane.RT-PCR showed that the expression of NR1 subunit mRNA of the TG in the neonatal rat hippocampus was significantly lower than that of the CG (0.3811±0.1043,0.9318±0.1593 repectively,P<0.05).Conclusion:Passive smoking during pregnancy can influence the ultrastructure of the CA1 regions and reduce the expression of NMDA receptor NR1 subunit mRNA in the neonatal rats hippocampus,which probably is one of the mechanisms to impair the neural functions.
