应用RAPD标记检测小豆种质资源的遗传多样性初探
Genetic Diversity of Adzuki Bean (Vigna angularis) Germplasm Resources Based on RAPD Markers
【摘要】 检测野生型、半野生型及栽培型小豆种质资源的遗传多样性 ,对进一步探讨小豆的起源地、品种分化和传播途径具有极其重要的价值。本研究用RAPD标记方法对来自中国、日本、韩国、不丹、尼泊尔、越南等 6国 99份小豆种质资源DNA的遗传多样性进行了检测。 7个引物共检测到 10 2条带 ,具有多态性的谱带 10 0条 ,其中 7条带仅存在于单个材料中。利用这些多态性谱带数据进行聚类分析 ,可明显划分出 5大类群。中国的小豆种质资源遗传多样性最为丰富 ,在5个类群中都能找到 ,其次为日本的小豆种质资源。韩国、越南、不丹、尼泊尔小豆种质资源遗传多样性相对较差 ,其栽培型与野生型、半野生型的遗传亲缘关系较近。我国华北地区小豆育成品种与东北地方品种的亲缘关系最近。
【Abstract】 It is very important to detect the genetic diversity of wild, weedy and cultivated adzuki beans for further researching the process of diffusion, evolution and the origin of adzuki bean germplasm resources. RAPD (Random Amplified Polymorphic DNA) variation was assessed in 99 materials of adzuki bean including the three species from China, Japan, Korea, Bhutan, Nepal and Vietnam. 102 bands including 100 polymorphic bands were detected by 7 RAPD primers. Seven bands were uniquely observed in single material. Five groups were clustered based on those data of polymorphic bands. Chinese accessions, observed in all 5 groups, exhibited the highest genetic diversity, and then the Japanese material. (Rela)tively low variation was observed in the material from Korea, Bhutan, Nepal and Vietnam. Close genetic relationship among wild, weedy and cultivated adzuki bean was also observed in the materials from the four countries. The cultivars race of northern China had the closest genetic relationship with the local cultivars of northeast China.
