中药防治硫酸镍致大鼠睾丸组织损伤的实验研究
Protective effect of Chinese herbal drug on testis’ damage induced by nickel sulfate(NiSO4)
【摘要】 目的 :探讨中药扶正解毒汤 (FJD)防治硫酸镍致睾丸组织损伤的作用机制。方法 :6 0只Wister大鼠随机分为 6组 :空白对照组、NiSO4染毒组、FJD不同剂量治疗组和FJD不同剂量预防组。中药治疗 4周后 ,测定精子细胞中Ca2 + ,睾丸组织中丙二醛(Maleicdialdehyde ,MDA)、超氧化物歧化酶 (Superoxidedismutase ,SOD)含量、以及谷胱甘肽过氧化物酶 (GlutathionePeroxidase ,GSH -PX)活力。结果 :染毒组Ca2 + 、MDA含量升高 ,SOD含量和GSH -PX活力下降 ,与对照组比较P <0 .0 5 ;中药治疗后各组Ca2 + 、MDA含量降低 ,SOD含量和GSH -PX活力升高 ,与染毒组比较P <0 .0 5。结论 :硫酸镍可导致大鼠精子细胞钙超载 ,睾丸组织脂质过氧化损伤 ,FJD具有调节钙稳态和抗睾丸组织脂质过氧化损伤的作用。
【Abstract】 Objective:To inquire into the mechanism of prevention and cure effect on testis’ damage induced by nickel sulfate(NiSO 4) with Fuzheng Jiedu Tang(FJD). [WT5HZ]Methods:Sixty rats were divided into 6 groups: the control , nickel sulfate(NiSO 4) group, FJD treated group and FJD prevention groups with different doses. After treated for 4 weeks,Ca 2+ in spermatid, the contents of MDA and SOD,GSH-PX vigor in constitution were estimated. [WT5HZ]Results:Ca 2+ and MDA content was increased, SOD content and GSH-PX vigor were decreased in NiSO 4 group compared with that of the control(P<0.05); After treatment with FJD, Ca 2+,the contents of MDA,SOD and GSH-PX vigor were improved(P< 0.05).Conclusion:Nickel sulfate could make calcium overload in spermatid and testis’ lipid peroxidation in rats, FJD could recover the changes of Ca 2+, MDA, SOD and GSH-PX vigor induced by nickel sulfate and effectively prevent testis’ damage.
