两种小鼠肺纤维化造模方法的比较

【摘要】 目的 对比研究肺纤维化造模方法 ,探寻出一种较为简便可靠的方法。方法 将雾化吸入不同剂量博莱霉素处理的小鼠及进行气管给药的小鼠 ,分别于 7、14、2 8d处死后 ,取其肺叶制备病理切片 ,染色后镜检。结果 一定时间的博莱霉素雾化吸入给药及气管给药 ,均可造成肺纤维化模型。前者药物在肺内分布均匀 ,而后者分布不均。结论 气管内直接注入药物的造模方法由于药物入肺分布不均 ,造成各肺叶纤维化程度有明显差异 ;雾化吸入法操作简便 ,吸入药物在肺内分布均匀 ,是一种较为理想实用的肺纤维化造模方法。

【Abstract】 Objective To seek a convenient and reliable model of pulmonary fibrosis, provide a better model for future pulmonary fibrosis research, and improve the level of pulmonary fibrosis management. Methods The mice were administerted with aerosol inhalation of bleomycin (BLM) or by the trachea, and were sacrificed after treatment on days 7, 14 and 28, respectively, and their lungs were harvested for histological studies. Results Both of the methods could induce pulmonary fibrosis. BLM was distributed unevenly by tracheal inhalation, while BLM was distributed evenly by aerosol inhalation for acertain period of time. Conclusion Because of uneven distribution of drug in the lung, the way by trachea will cause obvious difference in degree of pulmonary fibrosis, aerosol inhalation will bring a better result because of the convenient operation and even distribution of inhaled drug in the lung.
