一种简单实用的谷氨酸浓度测定法
A Simple Method in Measuring the L-Glutamic Acid Concentration
【摘要】 谷氨酸和茚三酮可以发生特有的显色反应 ,在 pH 5~ 6时产物蓝紫色最明显。其颜色深浅与谷氨酸浓度大小有关。用分光光度计可以在其最大光吸收波长处测定其OD值。分别以OD值和相应的一系列浓度的L谷氨酸纯品浓度值为纵坐标和横坐标 ,绘制出 1条标准曲线。谷氨酸发酵液中的谷氨酸含量值即可通过分光光度计测得它与茚三酮在最适 pH处反应得到的蓝紫色产物的OD值 ,然后从上述标准曲线上查到。误差率在 1 0 %左右。这为谷氨酸定量测定提供了一种简单实用的方法。
【Abstract】 L-Glutamic acid can specifically react with Ninhydrin especially in the condition of pH5~6.The reactive product shows bright blue and purple.By using the spectrophotometer we can mensur its OD value.Set a series of OD value as the X-coordinate and its corresponding concentrations of pure L-Glutamic acid as the Y-coordinate,then we can work out a standard curve.After reacting with Ninhydrin in the optimum pH 5~6 the fermentation product can be measured its OD value by the spectrophotometer.From the standard curve can we catch its correstponding concentration.The error rate of this method is about 10%.It offers a simple and practical measuring method for L-Glu.
