醋酸铅体外染毒对移植后小鼠胚泡发育的影响

【摘要】 目的 用胚胎移植技术评价醋酸铅对啮齿类动物的生殖毒性。方法 采用 2 0和 40 μg mL浓度的醋酸铅对小鼠着床前胚胎进行体外染毒 ,将染毒后可胚泡化的胚胎移植于假孕母鼠体内继续生长发育 ,观察出生前胎鼠形态学改变和出生后子鼠的行为改变 ,并对子鼠大脑进行病理检查。结果  ( 1)在实验剂量下 ,各染毒组出生前胎鼠形态学检查未见异常。 ( 2 ) 40 μg mL醋酸铅染毒组子鼠出现行为异常 ;回旋运动、阴性趋地运动、翻正反射和断崖回避阳性日均迟于阴性对照组 (P <0 0 5、P <0 0 1、P <0 0 5、P <0 0 5 ) ;悬挂时间明显短于阴性对照组 (P <0 0 5 ) ;10日龄游泳积分低于阴性对照组 (P <0 0 1)。 ( 3 )电镜超微结构显示各染毒组子鼠大脑中毒性损伤。结论 小鼠植入前胚泡体外醋酸铅染毒影响移植子鼠的行为功能 ,并引起中枢神经组织病理损伤 ;胚胎移植技术用于检测化学物胚胎毒性 ,排除母体因素的干扰 ,是一重要研究手段

【Abstract】 Objective Evaluate the reproduction toxicity of lead acetate to rodent with the embryos transplant technology.Methods The preimplantation embryos of mice were exposed to lead acetate in vitro. After exposure,the chemically treated blastocysts were transferred to the pseudopregnant surrogate mothers.We have investigated the morphologic changes of the litters before birth and the behavioral function of offspring after birth.The histopathology of the cerebral of offspring were also observed with electron microscope.Results (1)There were no gross dysmorphogenic effects in the litters when exposed to the doses of the experiment.(2)When exposed to 40 μg/mL lead acetate,the offspring showed behavior abnormalities.(3) The examination of ultrastructure of electron microscope showed poisonous damage in the offspring.Conclusion The preimplantation embryos of mice exposed to lead acetate in vitro effected the transferred offsprings,and damaged central nerve tissues.The embryos transplant technology is a valuable methods to detect the embryo toxicity of chemicals,and in this way the effect of the matrix can be avoided.
