奥曲肽对胃癌细胞系生长和胰岛素样生长因子-Ⅰ、Ⅱ含量的影响
Influences of Octreotide on the Growth of Gastric Cancer Cell Line and the Contents of Insulin-like Growth Factor-Ⅰ and Ⅱ
【摘要】 背景:生长抑素类似物奥曲肽对上皮细胞的增殖和胃肠激素的分泌具有抑制作用,并能抑制某些肿瘤的生长。目的:研究奥曲肽对胃癌细胞系体外生长的抑制作用和对胰岛素样生长因子(IGF)鄄Ⅰ、IGF鄄Ⅱ含量的影响。方法:应用四甲基偶氮唑蓝(MTT)比色法检测经奥曲肽处理的人胃癌细胞系SGC7901的生长情况,并以放射免疫测定检测SGC7901细胞培养液中IGF鄄Ⅰ、IGF鄄Ⅱ的含量。结果:0.01、0.1、1.0和10.0μg/ml奥曲肽对SGC7901细胞的生长均有抑制作用(与阴性对照组比较,P<0.01),抑制率分别为7.5%、11.2%、20.8%和19.9%,其抑制作用具有良好的量效关系和饱和现象。经0.1和1.0μg/ml奥曲肽处理的SGC7901细胞,培养液中IGF鄄Ⅰ、IGF鄄Ⅱ的含量显著低于阴性对照组(P<0.01)。结论:奥曲肽能显著抑制胃癌细胞系SGC7901的体外生长,下调细胞培养液中IGF鄄Ⅰ、IGF鄄Ⅱ的含量。奥曲肽可能通过抑制IGF鄄Ⅰ、IGF鄄Ⅱ的分泌而发挥抑制胃癌生长的作用。
【Abstract】 Background: Octreotide, a somatostatin analogue, has been shown to have antiproliferative effect on the epithelial cells and can suppress the secretion of gut hormones. Moreover, it can inhibit the growth of some kinds of tumors. Aims: To investigate the inhibitory effect of octreotide on the growth of gastric cancer cell line in vitro and its influence on the contents of insulin-like growth factor (IGF)-Ⅰ and Ⅱ. Methods: The proliferation of human gastric cancer cell line SGC7901 treated with octreotide was determined by methyl thiazolyl tetrazolium (MTT) chromatometry. The contents of IGF-Ⅰ and IGF-Ⅱ in the culture media of SGC7901 cells were measured by radioimmunoassay. Results: The growth of SGC7901 cells treated with octreotide was obviously inhibited as compared with that of the negative controls (P<0.01), the inhibition rates were 7.5%, 11.2%, 20.8% and 19.9% when treated with 0.01, 0.1, 1.0 and 10.0 μg/ml octreotide, respect-ively. The inhibitory effect of octreotide occurred in a dose-dependent manner and was saturated at the dosage of 1.0 μg/ml. Meanwhile, the contents of IGF-Ⅰ and IGF-Ⅱ in the culture media of SGC7901 cells were significantly decreased in 0.1 and 1.0 μg/ml octreotide-treated subgroups as compared with those in the negative controls (P<0.01). Conclusions: Octreotide can significantly inhibit the growth of gastric cancer cell line SGC7901 in vitro, and down-regulate the contents of IGF-Ⅰ and IGF-Ⅱ in the culture media. The inhibitory effect of octreotide on gastric cancers may be through the suppression of IGF-Ⅰ and IGF-Ⅱ secretion.
