吐温80-硫酸钠液—固萃取分离头孢拉定
THEPARTITION OF CEPHALOSRINS BY TWEEN80-NA2SO4 LIQUID-SOLID EXTRACTION SYYTEM
【摘要】 对头孢菌素类抗生素———头孢拉定在吐温80-硫酸钠液—固萃取体系中的分配行为做了初步研究。试验了成相聚合物吐温80浓度,成相盐硫酸钠浓度和体系pH对头孢拉定在吐温80固相中正萃取的影响,硫酸钠浓度对其在硫酸钠盐水相中反萃取的影响。结果表明:在吐温80在体系中的浓度为12%,硫酸钠浓度为1 48mol/L,pH7 5时,吐温80固相对头孢拉定正萃取率为58 2%;调整二次成相硫酸钠浓度0 63mol/L,pH3 5时,硫酸钠盐水相对头孢拉定反萃取率近95%。结果表明,头孢拉定在该体系中有较高的萃取率和较好的稳定性,显示吐温80-盐液—固萃取体系应用于分离抗生素等生物小分子有较大开发价值。
【Abstract】 For the first time, the distribution of small biomolecular, cefradine, in the polymer-salt aqueous liquid-solid extraction system of Tween80-Na2SO4 was investigated. Some parameters contributed to the partitioning behavior of cephalosporins, for exampe the concentration of the phase-forming polymer,Tween80, the phase-forming slat Na2SO4and the pH of the system were studied. The optimal conditions were obtained. For cefradine, the concentration of Tween80 was 12%(volume fraction), Na2SO4was 1.48mol/L, and with pH7.5,the recovery of cefradine in Tween80 phase was 58.2%.For the concentration of phase-forming salt,Na2SO4,adjusted to 0.63mol/L, and with pH3.5,the dissolved Tween80 re-formed liquid-solid two phase system, the cefradine was extracted to the salt aqueous with recovery nearly 95%.The result implied that the extraction system had no damages to the bioactivity of cefradine. From the work proceeded, we can draw a conclusion that the polymer-salt liquid-solid extraction system could be applied for the partitioning of small biomoleculars such as cepholosporins with satisfied recovery and imperceptible damages to the bioactivities of the biomoleculars.
