以RAPD方法分析岩原鲤分类地位
Indentification Analysis of Procypris rabaudi by RAPD
【摘要】 采用opaopoopq等系列44个随机引物对鲤亚科与亚科的几个种类进行随机扩增多态DNA(RAPD)扩增,通过对电泳结果进行系统分析,得出各种间遗传距离分支图.按节点的遗传距离最大数值0.232可以分出亲缘关系最远的两个群体,即由两种普通鲤为一群体,以及由岩原鲤、倒刺和白甲鱼组成的另一群体.从节点间遗传距离数值叠加计算可以得出岩原鲤与其他各种鱼类亲缘关系,遗传距离由近到远分别是:岩原鲤与倒刺(0.344)<岩原鲤与白甲鱼(0.388)<岩原鲤与普通鲤(0.443),岩原鲤与倒刺的遗传距离最小,这一结果表明岩原鲤更偏向于亚科,而不是鲤亚科.
【Abstract】 Cyprinidae is the largest and most diversity taxa of freshwater fishes in China. Although systematic and phylogenetic descriptions of the family have been done, there are still some disagreements about taxonomy. For example, Cyprininae and Barbinae are morphologically close to each other. Cyprininae is regarded as distinct from Barbinae and it can be separated from latter by its serrated anal spine. However, some researchers are doubtful about this morphological classification criterion. Therefore, the systematic description of some fishes in the two subfamilies is better to be completed by adding some molecular evidence. Procypris rabaudi, is a kind of fish to be revalued. RAPD method was performed to address the question. Cyprinus carpio xiangjiangnensis and Cyprinus carpio singuonensis were chosen as representatives of Cyprinidae, while Spinibarbus sinensis and Onychostoma sima were chose as representatives of Barbinae. 44 random primers were used to amplify the five species of fishes. Phylogeny analyses of RAPD results yielded two monophyletic clades: two xommon carps and other species. The genetic distance of five species of fishes by RAPDIST software showed the relative relationships: Procypris rabaudi-Spinibarbus sinensis ((0.344)<Procypris) rabaudi-Onychostoma simus ((0.388)<Procypris) rabaudi-Cyprinus c. xiangjiangnensis+ Cyprinus c. singuonensis (0.443). The results suggested that Procypris rabaudi is closer to Barbinae than Cyprininae.
