【摘要】 目的　探讨幽门螺杆菌 (HelicobacterpyloriH .pylori)鞭毛基因 (flaA和flaB)PCR RFLP分型及其与胃十二指肠疾病的关系。方法　用PCR技术分别扩增 46株从消化性溃疡 ( 2 5株 )和非消化性溃疡 ( 2 1株 )患者胃黏膜中分离得到的H .pyloriflaA和flaB基因 ,并用HpaⅡ和HindⅢ消化PCR扩增产物 ,琼脂糖凝胶电泳分离酶切片段 ,将酶切片段编码后输入SPSS10 .0软件进行聚类分析 ,并比较不同的基因型别在溃疡组和非溃疡组的分布。结果　 46株H .pylori菌株flaA基因HpaⅡ和HindⅢ酶切后分别产生 4种和 3种酶切图谱 ;flaB基因HpaⅡ和HindⅢ酶切后分别产生 1种和 2种酶切图谱。综合flaA和flaB基因酶切结果 ,46株菌株可分为四个基因型别。但这四个基因型别在溃疡组和非溃疡组的分布差异无统计学意义 (P =0 .5 13 )。结论　应用PCR RFLP方法可以将H .pylorifla基因分为不同的基因型别 ,但胃十二指肠溃疡和非溃疡组的菌株fla基因型别无差别更多还原

【Abstract】 Objective To investigate PCR-RFLPs for H.pylori fla gene and their relationships with gastroduodenal diseases.Methods flaA and flaB genes of 46 H.pylori from patients with peptic ulcer(25 isolates)and non-peptic ulcer(21 isolates)were amplified by PCR.The PCR products were digested with HpaⅡ or HindⅢ.The PCR-RFLP patterns were coded with different bands and analyzed by cluster analysis with SPSS 10.0 software.Compared the distribution of different genotypes in peptic ulcer and non-peptic ulcer.Results There were 4 restriction patterns in PCR products of flaA gene for 46 strains of H.pylori,digested with HpaⅡ,and 3 patterns with HindⅢ.There was only one restriction pattern in PCR products of flaB gene for 46 strains of H.pylori,digested with HpaⅡ,2 patterns with HindⅢ.46 clinical strains were classified into four groups by cluster analysis,when the patterns of flaA gene were combined with flaB gene.The relationships between the different genotypes and the determined diseases were not found(p=0.513).Conclusion The fla genes could be classified into several genotypes,but are conserved in clinical isolates.更多还原
