RNA干涉及其应用
RNA Interference and Its Applications
【摘要】 RNA干涉(RNAinterference,RNAi)是生物体内的一种通过双链RNA(dsRNA)来抵抗病毒入侵和抑制转座子活动的自然机制,它是转录后水平基因沉默(PTGS)的一种。RNAi的分子机制可分为三个阶段siRNA形成阶段、扩增阶段和效应阶段。RNAi作为基因沉默的有效工具,已被广泛应用于功能基因组学研究、基因治疗和新药的研究与开发等方面。
【Abstract】 RNA interference (RNAi, China) is a natural mechanism in organisms in resistance to virus invasion and inhibition of transposon mobility by double-stranded RNA(dsDNA, China). It is a kind of post- transcriptional gene silencing (PTGS, China). RNAi exists in a wide variety of organisms. RNAi molecular mechanism includes periods of initiation, amplification, and effect . As a gene silencing tool, RNAi has been widely used in functional genomics research, gene therapy, and new drug research and development.
