利用牛血块分离提取铜锌超氧化物歧化酶的工艺研究
THE TECHNICAL RESEARCH OF EXTRACTING COPPER AND ZINC SUPEROXIDE DISMUTASE(Cu,Zn-SOD) FROM BOVINE BLOOD
【摘要】 改进了传统提取超氧化物歧化酶的工艺 ,即直接用血块 ,加入保护剂、溶膜剂 ,采用机械破碎法破膜 ,热变性及两次乙醇 -氯仿沉淀除杂蛋白 ,然后丙酮沉淀 ,按照此工艺分离提取获得高纯度的Cu ,Zn SOD ,比活达到 6 988U/mg·pro。
【Abstract】 The traditional technology of producing SOD has been improved by us.That is,Higher purity Cu,Zn-SOD is extracted by added bovine blood directly,added protective agent and detergent,mechanical cracked for membrane is adopted,thermal denaturalization,twice ethanol-chloroform are used to remove impurity protein,then acetone precipitation.Its specific activity reach 6988 U/mg·pro.
