灵芝醇溶酸性组分的抗肿瘤作用

【摘要】 本文探讨了灵芝醇溶酸性组分 (ethanol solubleandacidiccomponents,ESAC)的抗肿瘤作用。用溶剂提取法从灵芝精粉中提取ESAC并利用高效液相色谱对其主要成分进行分析。利用MTT法检测ESAC对 3种细胞系 (人肝癌系BEL74 0 2、人宫颈上皮癌细胞系HeLa和人脐静脉血管内皮细胞系HUVEC)体外生长的影响。同时通过腹腔注射给药测定ESAC对BEL74 0 2瘤株在裸鼠皮下生长的影响。结果显示 ,ESAC对 3种细胞的毒性有较大差异 ,IC50 值分别为 12 .4 μg/mL(BEL 74 0 2 )、12 9.2 μg/mL(HeLa)和 5 2 6 .6 μg/mL(HU VEC)。动物实验表明 ,2 0mg/kg/dESAC组的肿瘤抑制率为 4 3.9% ,而 5mg/kg/d组的肿瘤抑制率高达 74 .9% ,且毒副作用较小。本文结果提示 ,灵芝ESAC组分对肝癌生长有一定的选择性抑制作用。

【Abstract】 The effects of ethanol-soluble and acidic components (ESAC) from Ganoderma lucidum on tumor growth were investigated in this paper. EASC was prepared by solvent extract method and its components were analyzed by high performance liquid chromatography (HPLC). Effects of ESAC on in vitro growth of 3 cell lines (including human hepatoma BEL 7402,Human cervix uteri tumor-derived HeLa and human umbilical vein endothelial cell line HUVEC) were determined by MTT method. Meanwhile,ESAC was administered to nude mice by peritoneal injection to examine the effects of ESAC on in vivo growth of BEL 7402 hepatoma. In vitro experiments showed IC 50 values of ESAC towards 3 cell lines were 12.4 μg/mL(BEL 7402),129.2 μg/mL(HeLa) and 526.6 μg/mL (HUVEC),respectively. Data from animal tests indicated the tumor inhibition ratios of ESAC were 43.9% (20 mg/kg/d) and 74.9% (5 mg/kg/d) with low toxicity. The results indicated ESAC from G. lucidum had the ability of inhibiting the hepatoma growth selectively,and therefore was a potential product.
