一氧化氮和内皮素在无心跳供肝缺血再灌注损伤中的作用
Study on the Role of Nitric Oxide and Endothelin in Ischemia-Reperfusion Injury of Non-Heart-Beating Liver Graft
【摘要】 目的 :通过大鼠原位肝移植模型 ,观察心脏停搏后不同时间供肝肝移植后的一氧化氮 (NO)和内皮素 (en dothelin ,ET)的水平及其和缺血再灌注损伤的关系。方法 :3个实验组分别取心脏停搏 0 ,30 ,4 5min后供肝行原位肝移植术 ,检测门静脉复流后不同时间点的肝功能、血中的肿瘤坏死因子α(TNF α)、NO和ET水平及肝脏形态学观察。结果 :3个实验组与对照组及相互之间门静脉复流后不同时间点的肝功能、TNF α、NO和ET水平均有显著性差异(P <0 .0 5 )。结论 :NO产生减少 ,ET的产生增多 ,NO ET的失衡是造成无心跳供肝缺血再灌注损伤的主要原因之一。
【Abstract】 Objectives: Rats orthotopic liver transplantation model were used to observe the level of nitric oxide (NO) and endothelin (ET) and the relation between NO, ET and liver graft ischemia-reperfusion (IR) injury at different time after cardiac arrest.Methods: Orthotopic liver transplantations were proceeded at 0, 30, and 60 min after cardiac arrest in three experimental groups. The levels of NO, TNF-α, ET, liver function test and histopathological examination of postreperfusion liver biopsies were all analyzed. Results: The liver functions of three experimental groups and control at different time after portal vein reflowing, the levels of NO, the levels of ET, and TNF-α were significantly different in all groups (P<0.05). Conclusion: The reduction of NO, the increase of ET and the unbalance between NO and ET play important roles in the development of IR injury of liver graft from NHBDs.
