人骨形成蛋白2和地塞米松对不同来源成骨细胞的作用
The Effects of BMP-2 and Dexamethasone on Different Kinds of Osteoblasts
【摘要】 目的 :观察不同浓度的人骨形成蛋白2(BMP2)和地塞米松 (DEX)对3种来源成骨细胞的作用 ,并比较其特性。方法 :体外培养胰酶消化法所得的正常成人的成骨细胞、成骨样细胞系和由骨髓单个核细胞 (MNC)经DEX诱导7d后所得的成骨细胞 ,经不同浓度的BMP2和DEX处理 ,观察细胞形态 ,并进行MTT试验和测定细胞裂解液的碱性磷酸酶 (ALP)。结果 :对正常成人的成骨细胞 ,BMP2处理48h ,72h对第4代和第8代的细胞无明显促增殖和分化作用 ;DEX作用48h和72h促第4代细胞增殖且表达ALP明显增加 ,DEX浓度为10-8 mol/L时其促增殖和分化的效果最佳 ,DEX对第8代细胞无明显作用。对成骨样细胞系BMP2促增殖作用不明显 ,DEX处理48 ,72h后明显促其增殖 ;BMP2及DEX作用72h后ALP表达明显增加 ,当作用于源自骨髓MNC的成骨细胞48 ,72h均促其增殖和分化。结论 :BMP2和DEX对不同成骨细胞的效应因细胞来源和作用时间不同而有差别
【Abstract】 Objective:To observe the effects of different concentrations of bone morphogenetic protein2(BMP2)and dexamethasone(DEX)on three kinds of osteoblasts and compare their characters.Methods:Three kinds of osteoblasts were cultured in vitro which included human adult osteoblasts isolated from trabecular bone with trypsin digestion, osteoblastlike cell line,and osteoblasts induced by bone marrow monocytes by DEX for 7 days.Osteoblasts were treated with different concentrations of BMP2 and DEX.Morphological changes were investigated.Proliferations were measured with methylthiazol tetrazlium(MTT)method,and alkaline activity(ALP)in the lysate was determined.Results:For the normal adult osteoblasts,BMP2 had no significant effects on the proliferation and differentiation of the fourth and eighth passage cells after 48 and 72 h.DEX could improve the fourth cell’s growth,and significantly increased the expression of ALP after 48 and 72 h.DEX had a significant effect under the condition of 10-8 mol/L.For osteoblastlike cell line,BMP2 had no significant effect on its proliferation and the effect of proliferation was significant with the use of DEX after 48,72 h.BMP2 and DEX could enhance ALP activity after 72 h.For the bone marrowderived osteoblasts,BMP2 and DEX significantly increased the proliferation and differentiation after 48 h and 72 h.Conclusion: The effects of BMP-2 and DEX on different osteoblasts varied because of the different sources and exposure duration.
