癫痫大鼠海马白细胞介素6免疫反应性的变化

【摘要】 目的 探求白细胞介素 6 (IL 6 )与癫痫发病之间的关系。方法 将 6 1只Wistar大鼠分成 3组 ,实验组侧脑室注入马桑内酯 (CL) ,2 μl(0 0 5mg/kg体重 ) ,生理盐水对照组注入等量的生理盐水 ,空白对照组不做任何处理。实验中对大鼠的行为及脑电图 (EEG)进行观察和记录 ;同时运用免疫组织化学方法 ,对大鼠海马IL 6的免疫反应性进行分析。结果 实验组大鼠侧脑室注射CL过程中即诱发癫痫 ,且测到痫性脑电图 ,生理盐水对照组、空白对照组无癫痫症状及癫痫样脑电图出现 ;实验组大鼠海马CA1、CA2 区IL 6免疫反应性较生理盐水对照组及空白对照组显著增强 (P <0 0 1) ,表现为海马CA1、CA2 区阳性反应细胞数目增多 ,着色明显加深 ;生理盐水组和空白对照组之间差异无显著性意义 (P >0 0 5 )。结论 癫痫的发病过程中海马神经元内的IL 6含量有显著的变化 ,二者之间可能存在着密切的联系。

【Abstract】 Objective To evaluate the relationship between cytokine interleukin-6 and epilepsy. Methods Sixty-one Wistar rats were divided into three groups: one experimental group and two control groups including a saline and a blank one. Two l (0.05 mg/kg weight) coriaria lactone (CL) and the same amount of saline were injected into the lateral ventricles of the rats in the experimental group and the saline group respectively, while nothing done to the blank group. The behavior and eletroencephalogram (EEG) was observed and recorded. Immunohistochemistry was used to monitor the interleukin-6 immunoreactivity in the hippocampus of all rats. Results In the experimental group, during injection of CL into the lateral ventricles of the rats, seizures were induced and the epileptic EEG recorded. The IL-6 immunorecativity of the CA1 and CA2 areas in hippocampus was stronger in experimental group than that of the two control groups (P<0.01), showing increased number of positive neurons, more processes and branches, and stronger staining. No difference was found between the two control groups. Conclusion The IL-6 content in the hippocampus changes greatly during the process of epilepsy, and, there exits a close relationship between IL-6 in hippocampus and epilepsy.
