细胞周期检测点激酶Chk1和Chk2在急性白血病骨髓单个核细胞中表达的研究

【摘要】 目的 探讨细胞周期检测点激酶 1,2 (Chk1,2 )在急性白血病中表达的意义。方法 用RT PCR方法、免疫组织化学方法及共聚焦显微镜检测Chk1和Chk2的mRNA和蛋白在白血病细胞株K5 6 2及急性白血病骨髓单个核细胞中的表达。结果 Chk1和Chk2的mRNA水平在急性白血病与增生性贫血之间无显著差异 ,Chk1和Chk2在K5 6 2细胞及白血病骨髓单个核细胞的细胞质和细胞核中均有表达 ,Chk1蛋白在初治及骨髓幼稚细胞大于 30 %的复治白血病的骨髓单个核细胞中核表达显著增高 ,Chk2蛋白在白血病骨髓单个核细胞中核表达与增生性贫血相比无显著差异。结论 Chk1和Chk2的mRNA水平与白血病发生无关 ,但Chk1蛋白表达水平与白血病发生有一定的相关性 ,Chk1有作为白血病治疗靶点的可能性 ,Chk2蛋白表达水平与白血病发生无关。

【Abstract】 Objective To investigate the role of Chk1 and Chk2 expression in the pathogenesis of leukemia. Methods K562 cells and bone marrow mononuclear cells (BMMNC) from acute leukemia and control were obtained. Semi quantitative RT PCR was used to detect Chk1 and Chk2 mRNA levels. Immunohistochemical analysis and confocal microscopy were used to detect the protein expression of Chk1 and Chk2. Results The level of Chk1 and Chk2 mRNA had no significant difference between the leukemia and control. Immunohistochemical analysis and confocal microscopy revealed that the expression of Chk1 and Chk2 was detectable in the nuclei and cytoplasm of K562 cells and BMMNC of leukemia and control. The expression of Chk1 in nuclei of BMMNC from leukemia was increased as compared with that from control. The expression of Chk2 had no significant difference between the leukemia and control. Conclusion The level of Chk1 and Chk2 mRNA had no relationship with the pathogenesis of leukemia. The expression of Chk1 protein in nuclei may take part in the pathogenesis of leukemia and Chk1 may be target of leukemia therapy.
