己酮可可碱对新生大鼠缺血缺氧性脑病的干预作用
Effects of pentoxifylline on newborn rats with hypoxic-ischemic encephalopathy
【摘要】 目的 :观察己酮可可碱 (pentoxifylline,PTX )对新生大鼠缺血缺氧性脑病 (hypoxic- ischemicencephalopathy,HIE)模型的干预作用。方法 :建立新生大鼠 HIE模型 ,72只出生 7d的 Wistar大鼠随机分为假手术组、HIE模型组和 PTX组 ,于缺血缺氧后 72 h取左脑 ,测定脑组织水含量、大脑皮层神经细胞内游离钙离子浓度、超氧化物歧化酶 (SOD)、丙二醛 (MDA)、一氧化氮合酶 (NOS)及一氧化氮 (NO)。结果 :与 HIE模型组相比 ,PTX组脑组织含水量、游离钙离子浓度和 MDA、NOS以及 NO水平显著降低 (P <0 .0 5、0 .0 1、0 .0 5、0 .0 5、0 .0 5 ) ,SOD水平显著升高 (P <0 .0 5 )。结论 :PTX对新生大鼠缺血缺氧后脑组织有保护作用 ,其机制可能与 PTX改善能量代谢及减少局部细胞毒性物质有关。
【Abstract】 Object: To observe the effects of pentoxifylline (PTX) on newborn rats with hypoxic-ischemic encephalopathy (HIE). Methods:Models of 7-day-old newborn Wistar rats with HIE were established and 72 rats were randomly divided into sham operation group, HIE model group and PTX group. The left side brains were taken out 72 h after hypoxic-ischemic operation. The contents of water, intracellular calcium and the levels of SOD, MDA, NOS, NO were measured. Results:Compared with HIE model group, the contents of water and calcium and the levels of MDA, NOS, NO were markedly reduced (P<0.05), while the level of SOD was increased significantly (P<0.05) for PTX group. Conclusion:PTX could protect brain of neonatal rat against hypoxic-ischemic damage, which might be the result of the improvement of energy metabolism and the removel of focal cytotoxic substence.
