琼扩试验检测鸡传染性法氏囊病卵黄抗体的研究
Agar Gel Immunodiffusion for Detection of IBDV Yolk Antibodies
【摘要】 采用琼脂扩散(AGP)试验检测免疫鸡血清和3种不同稀释液(氯仿、8%柠檬酸钠、16%氯化钠)处理的卵黄样品,结果表明,免疫后的卵黄样品与血清样品的AGP抗体效价一致,为1∶16。氯仿样品的检出率最高,为85%,血清样品的检出率为67%,8%柠檬酸钠和16%氯化钠样品的检出率分别为47%和25%。同时,对AGP检测结果分别在点样后24h、48h和72h进行观察,结果表明,AGP结果的最佳观察时间在24~48h。检测氯仿处理卵黄抗体的AGP可以代替检测血清抗体的AGP,以避免采血造成应激,具有重要的经济意义。
【Abstract】 The agar gel immunodiffusion (AGP) test has been taken for the detection of IBDV yolk antibodies, which were obtained by the treatment of egg yolk from immunized hens with 8% sodium citrate solution,16% sodium chloride solution and chloroform. The results show that there was no difference between yolk AGP antibody titres and serum AGP antibody titres , but the positive rates of sera and yolks treated with chloroform, 8% sodium citrate solution and 16% sodium chloride solution were 67%, 85%, 47% and 25%, respectively. The positive results of the AGP were observed after 24 hours, 48 hours, 72 hours respectively, and the best results were obtained from 24 to 48 hours. It is suggested that the gel test may be used for the detection of IBDV yolk antibodies instead of IBDV sera antibodies to avoid upsetting the chicken flock and it is of economic importance.
