HRP/PET自组装酶膜及其在光度分析中的应用
HRP/PET Self-assembled Film and Its Application in Spectrophotometric Determination of H2 O2
【摘要】 利用静电自组装技术在阴离子化的PET表面组装了单分子层的HRP酶膜 ,通过AFM对膜表面形貌进行了分析。制备的酶自组装膜在4℃的低温条件下 ,密封保存150d后 ,酶活性仍保留80 %以上。以微量比色皿为反应容器 ,考察了酶膜与H2O2 的显色反应动力学 ,该反应的表观米氏常数 Kmapp=3.2×10-5mol·L-1(相对于H2O2 底物) ,显色反应在5min内完成 ,对样品中H2O2 测定的回收率为96.5%~101.1 %。
【Abstract】 Monolayer HRP film was assembled on the surface of ionized PETusing electrostatic self-assembly technology.The topographies of the filmsurface were then examined byAFM.Itwas found thatthe relative acˉtivity of the self-assembled HRP filmstill retained82%after storing for150days at4℃.The kinetics of the color reaction of the enzyme filmwith H 2 O 2 in micro cuvette was determined using spectrophotometry.The color reaction was completed within5min and its apparent Michaelis constant( K m app )was3.2×10 -5 mol·L -1 .The recoveries for H 2 O 2 in rain water samples were96.5%~101.1%.
